Janus nanoparticles are nanomaterials with a unique asymmetric structure,which can integrate materials with two functions into a single particle to exert the synergistic effect of the two components in different application fields.Due to the multi-component and multifunctional properties of Janus nanoparticles,researchers have carried out extensive research on Janus nanoparticles.Recently,Janus nanoparticles have attracted extensive attention in the field of biomedicine,such as biosensing,drug delivery,bioimaging,and cancer combination therapy.In this study,we constructed a Janus nanocomposite structure by integrating mesoporous platinum(Pt)and mesoporous organosilica(JPMO-Pt),and explored their application in tumor therapy.The research content includes the following two parts:(1)Janus-structured mesoporous organosilica-mesoporous platinum nanoparticles(JPMO-Pt)-loaded halofuginone(HF)are used to achieve high-efficiency tumor therapy.HF not only inhibited tumor cell clones effectively,but also promoted the penetration of JPMO-Pt into tumor tissue to kill tumor cells efficiently by degrading various components in the tumor stroma,including collagen andα-smooth muscle actin.The results of in vitro and in vivo experiments showed that the JPMO-Pt-HF nanoplatform combined the photothermal effect of JPMO-Pt and the tumor cell inhibition and tumor matrix digestion effects of HF,which exhibited a good synergistic killing effect on tumor cells under laser irradiation.(2)The JPMO-Pt-CTPP-Zn Pc nanoplatform specifically targeting tumor cell mitochondria was constructed to realize tumor photodynamic therapy accordingly by modifying the mitochondriatargeting molecule carboxytriphenylphosphine(CTPP)on the surface of JPMO-Pt and loading the photosensitizer zinc phthalocyanine(Zn Pc).The prepared JPMO-Pt-CTPP-Zn Pc has the characteristics of uniform size,good dispersion and good biocompatibility.The in vitro results showed that JPMO-Pt-CTPP-Zn Pc had better mitochondrial targeting and stronger tumor cell killing effect than JPMO-Pt-Zn Pc.
