Malignant tumors can metastasize to distant sites through blood circulation or lymphatic circulation and develops advanced disease,which is also the main cause of death among patients with malignant tumors.Circulating Tumor Cells(CTCs)are tumor cells that fall off the surface of primary tumor tissue and enter the peripheral blood.It is formed by tumor cells with metastatic potential leaving the primary tumor site and entering the blood or lymphatic circulation,and forms new tumor lesions at a distance.Therefore,it plays an important role in the process of tumor metastasis.The detection of CTCs by peripheral blood has the characteristics of simple operation,high sensitivity and high specificity.Therefore,the detection of CTCs plays a very important role in the early detection and treatment of cancer,real-time disease monitoring,and monitoring of postoperative tumor patients and the evaluation of treatment effect.Organic Electrochemical Transistor(OECT)have attracted extensive attention in the field of biochemical sensing,due to their advantages of low cost,easy preparation,low operating voltage(<1V),strong Electrochemical activity,excellent environmental stability,and good biological compatibility.The circulating tumor cell sensor based on OECT has a very good application prospect with characteristics of fast detection speed and high detection sensitivity.Compared with other biosensors,the array organic electrochemical sensor has more advantages in detection and monitoring.It has the advantages of miniaturization and refinement.It can monitor stable current for a long time on the premise that each micro-sensor does not interact with each other,and obtain higher SIGNal-to-noise ratio of sensors with equal surface area,and can achieve lower current detection limit.The main research content of this paper revolves around how to prepare high sensitivity,high stability of organic electrochemical biosensors,in order to realize the low cost rapid realtime detection of circulating tumor cells,and explore the integration of equipment,which can provide theoretical and technical support for the establishment of a complete circulating tumor cell detection platform based on organic electrochemical sensors.The main work is as follows:1.Based on the basic theory and technology of biochemical sensing applications,Master the principle of OECT technology.Using poly3,4-ethylene dioxyethiophene/polystyrene sulfonate(PEDOT:PSS)conductive polymer as semiconductor film,Through the patterning of PEDOT:PSS films.Achieve OECT array sensor with high sensitivity and stability of common source drain and common gate modes.2.Based on the control theory and SCM technology,a multi-way selector is developed for the single module test and cycle test of the designed array organic electrochemical sensor.The multi-way selector can quickly locate the detection signal to the required detection part according to requirements.It can be matched with a variety of testing equipment,with independence and flexibility,be easy to operate,and will not have a negative impact on the test results.3.Use organic electrochemical sensors to modify biological protein,and achieve efficient capture of circulating tumor cells according to the specific antibody modification method.Achieve fast and efficient detection of circulating tumor cells by the use of semiconductor parameter analyzer and self-developed multiplex selector.
