| Tremella fuciformis and sialic acid both have the functions of maintaining beauty and keep young and antioxidation.Jiangxi is one of the main producing areas of Tremella fuciformis in China,Tremella fuciformis resources are quite rich,but there are almost no Tremella fuciformis deep-processed products.Sialic acid is mainly added to milk powder,bird’s nest and other products,and the product variety is relatively simple.This research uses Tremella fuciformis and sialic acid as the main raw materials to prepare sialic acid-Tremella fuciformis drink,which not only helps to promote the development of Jiangxi Tremella fuciformis industry,but also enriches the product variety of sialic acid and provides consumers with a new type of functional drink.The main results are as follows:(1)Using Tremella fuciformis and sialic acid as materials and pressure cooker as instrument to prepare sialic acid-Tremella fuciformis drink.Using single factor test and orthogonal test method,through sensory evaluation,the optimal craft formula of sialic acid-Tremella fuciformis drink was optimized as follows: the ratio of material to water is 1:10(weight ratio),the stewing time is 100 min,and the amount of white granulated sugar is 4%.The prepared sialic acid-Tremella fuciformis drink has good taste and flavor.(2)The main components and physical and chemical indexes of sialic acidTremella fuciformis drink were determined by high performance liquid chromatography,ultraviolet spectrophotometer and other instruments,the results showed that the content of sialic acid was 398.81 μg/g,the content of Tremella fuciformis polysaccharide was126.99 μg/m L,the content of fat was 0.0137 g/100 g,the p H was 3.87,the total content of dietary fiber was 0.0113 g/100 g,and the content of total protein was 0.0640 g/100 g,the content of ash was 0.0425 g/100 g,the content of water 95.10%,water activity0.9837,the content of solid was 41.32%,the content of soluble solid was 4.37 Brix%.(3)Through in vitro simulated digestion experiments,the scavenging rate of DPPH,ABTS and hydroxyl radicals during the sample digestion process was measured,and it was found that in the simulated digestion process in vitro,sialic acid can significantly improve the clearance rate of Tremella fuciformis drink to DPPH radicals,ABTS radicals and hydroxyl radicals,which means sialic acid can significantly enhance the in vitro antioxidant and anti-aging effects of Tremella fuciformis drinks.(4)The oxidative stress model was established by using menadione to induce zebrafish embryos to study the antioxidant effect and mechanism of beverages.By comparing different experimental groups of zebrafish embryos in vivo ROS quantity,the change of SOD activity and CAT activity and MDA content,it was found that sialic acid-Tremella fuciformis drink has significant inhibitory effect to generation of active oxygen induced by menadione,and can significantly enhance the activities of SOD enzymes and CAT enzymes in zebrafish,it can also reduce the content of MDA in zebrafish.The above experimental results all confirmed that sialic acid-Tremella fuciformis drink has strong antioxidant effect in zebrafish.(5)By measuring the content of sialic acid,Tremella fuciformis polysaccharide,p H,soluble solid content,total number of colonies and sensory scores in the sialic acidTremella fuciformis drink during storage,the storage stability of the sialic acidTremella fuciformis drink was studied,and the ASLT accelerated method was used to predict shelf life of sialic acid-Tremella fuciformis drink.The results showed that the shelf life of sialic acid-Tremella fuciformis drink under normal temperature(25℃)storage conditions was about 46 days,and the sialic acid content,Tremella fuciformis polysaccharide content,p H and soluble solid content in the sialic acid-Tremella fuciformis drink did not change significantly during the shelf life. |
