| In military operations,traffic accidents,and surgical operations,uncontrolled bleeding is the leading cause of death,so rapid hemostasis is necessary to improve survival.Currently commonly used hemostatic materials such as starch,gelatin,sponge and other hemostatic effects still need to be improved.In recent years,many studies have focused on hemostatic polymers,because the chemical structure of hemostatic polymers can be designed,and existing hemostatic substrates can be modified by immersing the polymer on the surface of the hemostatic material or by chemical grafting.The second chapter of this thesis synthesized PB with adjustable electropositivity by grafting β-propiolactone on polyamino polymer polydimethylaminoethyl methacrylate(PDM)and PB to prepare it into a hemostatic film and as a coating to prepare a modified medical gauze.The combination of PB2 and hydroxypropyl cellulose in an appropriate ratio has a procoagulant effect in vitro.The mechanism of action is that an appropriate amount of PB2 can promote the adhesion of platelets and has a procoagulant effect.It was further verified that the hemostatic film and modified gauze had a good hemostatic effect through the rat femoral artery injury hemostatic model.In the third chapter of this paper,a thermosensitive gel was constructed by compounding the preferred procoagulant polymer PB2 with poloxamer 407(F127),which was applied to wound hemostasis and vascular embolization.The hemostatic effect of hemostatic gel was verified by in vitro coagulation index,APTT,PT test and rat femoral artery injury hemostatic model,and the procoagulant effect of PB2 was further verified.The compound concentration of F127 and PB2 was adjusted,and the gel formulation was screened by rheological test and in vitro blood coagulation index for in vivo vascular embolization.Among them,F-PB(3,0.5)could effectively prolong the embolization time.In summary,a hemostatic polymer was synthesized in this paper,that is,a tertiary amine-based polymer was modified by adjusting the ratio of zwitterions and amino groups to make it have a coagulation-promoting effect.The compounding realizes the flexible application of coagulation-promoting ingredients.A simple and convenient preparation method of hemostatic material is provided,a new idea is provided for the research of polymer-based hemostatic material,and it has certain significance for the research and development of hemostatic material. |
