Fish swim bladder(flower gelatin)has been considered as a good medicinal material and tonic in traditional medicine in China and Southeast Asian countries,but as a waste product of modern processing of fish,it has not been reasonably and fully utilized,and its physiological efficacy has not been fully confirmed by modern scientific research,resulting in waste of resources and environmental pollution.In this study,investigated the aging-delaying effects and mechanisms of collagen peptides(CPs)using sturgeon swim bladder as raw material and Caenorhabditis elegans as a model,and developed the non-additive,highly active collagen peptides product to provide strong technical support for the scientific application of sturgeon swim bladder.The main contents and results are as follows:(1)The anti-aging effect of sturgeon swim bladder CPs was significantly higher than that of bovine skin,porcine skin,tilapia skin,cold water fish skin,sturgeon skin and cod swim bladder.The anti-aging effect of sturgeon swim bladder CPs prepared by trypsin method were significantly higher than gelatin,pepsin,simulated gastrointestinal digestion and free amino acids,indicating significant differences in the biological properties of CPs prepared from different biological sources and by different methods.Small molecule CPs(PP,AP,PGP,GPG)at a dose of 25 mg/m L inhibited the accumulation of lipofuscin(27.90%-38.18%),improved the body’s resistance to heat stress(11.45%-12.93%),oxidative stress(9.69%-38.27%)and hyperosmotic stress(38.49%-107.85%),delayed apoptosis(32.53%-48.35%)and enhanced barrier function(11.13%-25.24%).(2)PP up-regulated 252 and down-regulated 634 expressed genes in C.elegans,which were mostly associated with protein folding,significantly enriched in metabolic regulatory pathways,and prolonged the lifespan of C.elegans mainly through the inhibition of m TOR pathway.PGP up-regulated 1068 and down-regulated 1297 expressed genes in C.elegans,which mainly played important roles in response to external stress,bacterial and other biological stimuli and defense responses,and were mostly enriched in longevity regulation pathways,mainly through activation of multiple signaling pathways by overexpression of DAF-16,thus prolonging the lifespan of C.elegans.(3)Using blueberry juice as a carrier,two new collagen peptides products with high activity and no additives had been developed: Product A(blueberry juice 90.00%,sturgeon swim bladder CPs 10.00%);Product B(blueberry juice 84.27%,sturgeon swim bladder CPs 10.00%,pollen extract 5.00%,coniferous cherry extract 0.20%,elastin peptide 0.20%,sodium hyaluronate 0.20%,phytoselenoprotein 0.10%,rice extract 0.02%,zinc glycinate 0.01%).The mouse acute toxicity,heavy metal and microbiological tests of the two products all meet the relevant national standards,and the sensory analysis shows that the products have good comprehensive evaluation and consumer acceptance.
