Hemp seed oil(HSO seed oil,HSO)is a mixture of mixed glycerides extracted from hemp plant seeds,in which the content of polyunsaturated fatty acids(Polyunsaturated fatty acids,PUFA)is as high as 80% or more,with extremely high nutritional value Species biological activity.However,in the process of the extraction,processing,storage and application of HSO,PUFA are easily oxidized and deteriorated,which is a bottleneck in the development of seed oil application technology greatly limits the extraction method of HSO.This subject research establishes a new method of Pre-pressing-Enzyme-assisted Repressing(P-EAR)under low temperature conditions,and extracts HSO under optimized process parameters.The main physical,chemical indexes and ingredients of HSO were analyzed,and then HSO was used as the core material,egg phosphatidylcholine(EPC)and cholesterol(Cholesterol,Chol)were used as the wall materials.Ethanol injection-ultrasonic dispersion(EI-UD)is a new method.Under the conditions of optimized formulation and process parameters,liposomes encapsulating HSO were prepared under different storage conditions.And analyse the physical and chemical stability of liposomes.P-EAR method to extract HSO.Using hemp seed kernels as raw materials,a small screw press(Screw press,SP)is used for pre-pressing,and the pre-pressed cake is treated with Viscozyme L and Protex 6L enzyme preparations,and then the substrate is re-pressed.Prepress conditions: hemp seed kernel moisture 4.0%,pre-press temperature 45-55 ℃,press screw shaft speed 60 rpm,cake thickness 3.0 mm.After optimization by single factor test and response surface method,the optimal process conditions of enzyme-assisted process: enzyme addition amount 1.1%(w/w),p H value 10.0,time 2.8 h,temperature 52 ℃.Re-press conditions: press moisture 3%,temperature 45-55 ℃,press screw speed 60 rpm,cake thickness 1.0 mm.Store the pre-pressed oil and repressed oil at 4 ℃.The results of the comparative analysis show that the oil extraction efficiency of the control group is 71.8% and the total oil extraction efficiency of the P-EAR method is 80.5%;the HSO liposomes are prepared by the EI-UD method.First,using encapsulation efficiency(EE)and oil load rate(OLR)as response indexes,the orthogonal method was used to optimize the preparation conditions of liposomes,and liposomes encapsulating hemp seed oil were prepared by EI-UD method.Liposome characterization data show that the average particle size(Z-Ave)is122±1.64 nm,the polydispersity index(PD.I)is 0.21±0.06,the Zeta potential(ZP)is-36.5±0.32 m V,the encapsulation efficiency(EE)is 81.3±1.62% and the oil load factor(LOR)is 6.13 ± 0.07%.Transmission electron microscopy(TEM)and cryo-scanning electron microscopy(Cryo-SEM)observed that the liposomes showed a rounded shape;physical and chemical stability test conditions: 4 ℃ protected from light,25 ℃ protected from light and25 ℃ natural light,stored for 70 days.The analysis results show that the average particle size(Z-Ave)and polydispersity index(PD.I)increase with storage time,and they have temperature dependence and photosensitivity;Compared with the control group,liposome-encapsulated HSO.The degradation rate of medium total polyphenols(TPC)and Δ9-tetrahydrocannabinol(Δ9-THC)is effectively suppressed,and the ability to scavenge free radicals(DPPH·)remains good.Studies have shown that the P-EAR method has the characteristics of high oil extraction efficiency,low acid value and peroxide value of oil,high total tocopherol and cannabinoid content in the oil,which improves the oil production rate and oil quality for subsequent processing and application Created basic high-value conditions;HSO liposomes have high encapsulation efficiency,uniform particle size,and good physical and chemical stability.They have the protective effect of delivering HSO under sensitive conditions such as light and heat,and are expected to be used in precise and nutritious food.The processing strategy plays a unique role and expands a wider space for the application of HSO in multiple fields. |
