Advanced Glycation End Products(AGEs)are mainly a complex mixture formed by the Maillard reaction between aldehyde groups in reducing sugars such as fructose and free amino groups in proteins or DNA.Accumulation of AGEs in tissues and organs is closely related to some physical diseases such as diabetes,nephropathy and liver diseases.The current study of long-term intake of high AGEs diets has a particularly poor impact on liver health.To investigate the long-term effects of high AGEs diets on liver health and possible molecular mechanisms,this study used C57BL/6 mice as animal models to feed normal AGEs and heated high-AGEs diets for 12 months.This study analyzes the accumulation levels of Nε-carboxymethyllysine(CML),Nε-carboxyethyllysine(CEL),oxidative stress,inflammation,fibrosis,and AGEs receptor expression in liver tissue and by observing liver’s structure this study also tries to find out whether long-term intake of high AGEs diets damages the liver.Main conclusions are drawn as follow:(1)Heating significantly increases the content of CML,CEL and fluorescent AGEs in feed(p<0.01).Long-term intake of high AGEs diets significantly increases the mice’liver coefficient and significantly increases mice’serum transaminase levels(p<0.05).(2)Long-term feeding of high AGEs diets resulted in significant increases in CML and CEL levels in the mice’livers,and their levels gradually increases as the feeding time increases(p<0.05).Long-term intake of high AGEs diets induces the production of lipid peroxides such as malondialdehyde(MDA)and 8-isoprostaglandin in the liver of mice,and decreases total superoxide dismutase(T-SOD)activity.There is a time-dependent relationship(p<0.05).In addition,long-term intake of high AGEs feeds can cause significant browning in the liver of mice.Fatty liver degeneration occurs in the livers of mice fed normal diet,which however does not happen in the mice with high AGEs.(3)After long-term intake of high AGEs,RAGE mRNA and protein expression levels in the mice’livers increase significantly(p<0.05).In addition,long-term feeding of high AGEs diets significantly increases mRNA expression levels and protein secretion of inflammatory cytokines such as IL-6 and TNF-αin the liver(p<0.05),but it does not have obvious effects on livers’fibrosis-related factors(α-SMA and TGF-β1)protein levels(p>0.05).
