Diabetes mellitus is a common endocrine and metabolic disease characterized by hyperglycemia.It is mainly due to the lack of insulin secretion which will lead to elevated blood glucose and metabolic disorders in the body.For diabetic patients,their tissues such as kidneys,blood vessels,eyes,heart,and nerves are prone to experience dysfunction and chronic damage.In recent years,dietary supplementation of functional factors through intaking edible and homologous plants has become a new direction for the improvement of diabetes.Chinese wolfberry contains a variety of active ingredients,of which Lycium barbarum Polysaccharides is one of the important components,with functional activities such as anti-virus,anti-tumor,lowering blood pressure,lowering blood fat,improving memory.Cichoric acid is a phenolic acid Substances extracted from echinacea with the function of hypoglycemic action,anti-inflammatory effect,anti-obesity,anti-oxidation,and immunity enhancement.In this paper,the author used Poria and Echinacea as main raw materials to develop hypoglycemic beverage,conducting animal experiments on C57BL/6J male mice induced by streptozotocin(STZ).The details and results are as follows:(1)The selection of raw material of wolfberry and echinacea functional beverageIn order to maximize the acquisition of functional factors,a testing was conducted as follows: the same quality of wolfberry,wolfberry powder,echinacea purpurea flowers and its’ leaves were brewed and then their functional factor content was determined.The results showed that the extraction rate of Lycium barbarum Polysaccharides in the water extract of wolfberry powder was higher than that of wolfberry,and the content of cichoric acid in the water extract of Echinacea leaves was higher than that of Echinacea flowers.(2)The development of wolfberry and echinacea functional beverageThrough single-factor testing and orthogonal testing,the extraction formula of the water extracts from wolfberry powder,echinacea purpurea leaves was optimized.The optimization formula was as follows: as for the wolfberry powder,the brewing time of was 2.5 h,the amount was 25 g,the needed water is 1800 m L(in this condition,the content of polysaccharides was the highest);for the echinacea purpurea leaves,the time was 15 min,the amount was 2 g,and the needed water is 700 m L(In this condition,the content of cichoric acid was the highest).Based on sensory evaluation,the response surface methodology was used to optimize the formula of Echinacea purpurea beverage,determining the final ratio of wolfberry powder and Echinacea purpurea leaves water extracts was 7:3,the content of xylitol was 4%,and the content of citric acid was 0.04%.After testing and according to the national standard,indicators including physical and chemical properties and microbiological indicators were all within the qualified range.(3)The effect of wolfberry and echinacea functional beverage on hypoglycemicThe STZ was injected into the abdominal cavity of C57BL/6J mice multiple times to induce it to become a diabetic mouse.The formulated Echinacea purpurea drink,concentrated2 x Echinacea purpurea drink,and concentrated 4x enriched Echinacea purpurea drink were used as dietary supplements to investigate the drink’s hypoglycemic effect.Test results show that Echinacea Purpurea can effectively relieve mice’s weight loss,improve its’ insulin sensitivity,glucose sensitivity and,cognitive and working memory,as well as alleviate the damages of muscle tissue and pancreatic tissue of STZ-induced diabetic mice.The STZ was injected into the abdominal cavity of C57BL/6J mice multiple times to induce it to become a diabetic mouse.The formulated Echinacea purpurea drink,concentrated2 x Echinacea purpurea drink,and 4x enriched Echinacea purpurea drink were used as dietary supplements to investigate the drink’s Hypoglycemic effect,and the test results show that Echinacea Purpurea can effectively relieve weight loss in mice,improve insulin sensitivity,glucose sensitivity and cognitive and working memory in mice,as well as effectively relieve the damages of muscular tissue and pancreas of mice with STZ-induced diabetes.
