Actinidia Chinensis root have a good effect in traditional Chinese medicine,whether oral or topical,no toxicity,its pharmacological effects including:antioxidant effect,inhibition of tumor cells,the role of the immune activity,antiviral,anti-liver injury and antibacterial effect,Actinidia Chinensis root pharmacologically shows the above characteristics and its containing polysaccharide,an anthraquinone substance and saponins,particularlyis saponins substances.In this study,choose Qinmei Actinidia Chinensis root as raw material,I use ultrasonic technology to extract ursolic acid isolated from the root of the Actinidia Chinensis,and on the basis of single factor test,optimize ursolic acid of Actinidia Chinensis root ultrasonic extraction process,and then use Ursolic acid of extracted crude extracts for the study,to determination of its antioxidant properties,antibacterial properties the analytical,results are as follows:(1)In order to optimize ursolic acid of the Actinidia Chinensis root by the ultrasonic extraction process,on the basis of single-factor test,select the ethanol concentration,liquid ratio,extraction time,extraction temperature as the independent variable,ursolic acid extraction rate as a response,the use of center combination tests(Central Composite Design CCD)and response surface methodology to study each variable interaction ursolic acid yield,simulated quadratic polynomial regression equation prediction model,and to determine the optimal conditions of ultrasonic extraction process of ursolic acid:ethanol concentration of 73.8%,liquid ratio 7.80:1(V:W),extraction time 23.2 min,extraction temperature 79.1℃.This extraction conditions,ursolic acid yield(2.018 ±0.006)%,a better fit between the model predictions,and the purity of ursolic acid in the extraction liquid is 49.7%.(2)The ursolic acid crude extract had strong antioxidant capacity,diphenyl-2-acyl(DPPH·)radical,superoxide radical(O2-·),hydroxyl radical(·OH)have different degrees of scavenging effect and reducing power.·OH had a very strong scavenging effect,DPPH·had a strong scavenging effect,O2-·had a weak effect in scavenging.(3)Antibacterial activity of the ursolic acid crude extracts was studied by the plate method,the results showed that in the test within the concentration range for the selected E.coli,Staphylococcus aureus and Salmonella all have some antibacterial effect,among them,the inhibitory effect against Staphylococcus aureus is the strongest(in the test concentration,ursolic acid crude extract had inhibition effect when its concentration is 0.125mg/mL),followed by Escherichia coli(in the test concentration,ursolic acid crude extract had inhibition effect when its concentration is 0.25mg/mL),the inhibitory effect on Salmonella is poor(in the test concentration,ursolic acid crude extract had inhibition effect when its concentration isn 0.5mg/mL).
