Feline panleukopenia,also known as feline distemper,is a common infectious disease with high mortality and morbidity caused by Feline panleukopenia virus(FPV).Its clinical symptoms include high fever,vomiting and severe leukopenia,seriously endanger the health of domestic and wild cats.It has age susceptibility and seasonal susceptibility.kittens under 1 year of age that are not fully immunized are most vulnerable to FPV.FP occurs frequently in cattery and concentrated areas of domestic cats,which seriously endangers the health of domestic cats and wild cats.FPV belongs to Parvoviridae,Parvovirinae.VP2 protein is the main structural protein of FPV.It has multiple antigenic sites and can stimulate body to produce neutralizing antibodies.The purpose of this study is to develop a monoclonal antibody reagent that can be used for rapid diagnosis,epidemic monitoring or disease treatment of feline distemper,which has important significance for immune prevention,epidemiological investigation and diagnosis and treatment of cat distemper,and lays a foundation for the development of therapeutic monoclonal antibody against cat distemper.In this study,feline panleukopenia virus was the object of study.Monoclonal antibody was prepared after prokaryotic expression of its VP2 protein.Recombinant protein VP2 was used as immunogen and screening antigen.After four subclonal screenings,a hybridoma cell line 1C2 was obtained,which could secrete FPV VP2 monoclonal antibody specifically.Chromosome analysis and stability analysis showed that the cell line secreted antibody stably.The subtype of monoclonal antibody was identified as IgG1.Western Blotting results showed that the selected monoclonal antibodies could specifically bind to VP2 recombinant protein,which had potential application value.This monoclonal antibody lays a foundation for exploring the pathological mechanism,clinical detection and vaccine development of FPV.
