| SVB(Smaller with variable branches)was originally discovered in Arabidopsis and contains a DUF538 domain.The DUF538 family has been found to exist widely in monocotyledonous and dicotyledonous plants.Some studies have shown that this family is involved in multiple growth and development pathways of plants,but there is little research on its function in participating in non Biotic stress.This study used Arabidopsis as the material to preliminarily explore the function of At SVB2 gene in response to salt stress and ABA stress.The main results are as follows:1.The Arabidopsis genome encodes a total of 22 members of the DUF538 family,which only has one conserved domain of DUF538,including 6 SVBs.By analyzing the gene structure,it was found that 5 SVBs genes do not contain introns;Analysis of the protein structure revealed that the SVBs contains two motifs.The At SVB2 gene that this study focuses on is located on chromosome 1.2.Construct a plant overexpression vector 35S: HA-At SVB2,and obtain 8homozygous overexpression strains through identification.Using CRISPR/Cas9 gene editing technology,obtain two At SVB2 gene mutant strains were successfully.Purchased and screened one T-DNA insertion mutant of At SVB2 gene.3.Under salt stress,the transcription level of At SVB2 gene is down regulated;The germination rate and green seedling rate of the At SVB2 gene overexpression strain were higher than those of the wild-type;The germination rate of the mutant is lower than that of the wild type.4.Under ABA treatment,the mutant seeds of At SVB2 gene germinated faster than the wild-type and were not sensitive to ABA,while the overexpression plants of At SVB2 gene had similar germination conditions to the wild-type.Previous studies have shown that At SVB2 is an ABA responsive gene and plays a crucial role in trichome development.This study demonstrates that the At SVB2 gene is involved in the response of Arabidopsis to salt stress.It laid a foundation for studying the function of At SVB2 gene in non Biotic stress. |
