| Salinised soils can have a great impact on the ecology and agricultural production.Hundreds of millions of acres of salinised soils exist in areas such as the Songnen Plain.Therefore,how to develop and utilise these lands effectively has become one of the important tasks for ecological development and agricultural production in China.In this experiment,11 materials of Korean alkali fescue(Puccinellia chinampoensis Ohwi)were collected from the western region of Jilin Province,and their phenotypic traits were used for genetic diversity analysis;the seeds were subjected to salt stress germination experiments using different gradients of NaCl solution for a comprehensive analysis of seed germination;the seedlings were treated with semi-lethal concentration of NaCl solution The seedlings were treated with half lethal concentration of NaCl solution,and the physiological and biochemical indicators and plant height were used to analyse their salt tolerance,so as to screen out germplasm materials with excellent phenotypic traits and salt tolerance.The results of the study are as follows:(1)By analysing 11 phenotypic traits of wild Korean alkali fescue materials from different populations and comparing the phenotypic traits of each material,it was found that the intra-group variation coefficients of two materials,J10 and J09,were low,mostly around 15%,with a lower degree of variation and the most stable genetic degree compared with other materials.The genetic diversity indices of the populations were analysed and found to be in the range of 0.85-0.88 for all populations,with a high degree of heritability of traits in different materials,which is conducive to the selection of high-quality germplasm resources.By analysing the correlation of phenotypic traits of all materials,it was found that the seed yield of different Korean alkali fescue materials was positively correlated with the number of solid grains per spike and the weight of a single spike,and negatively correlated with the number of leaves per plant and the number of stalk nodes.(2)From the germination experiments on the salt tolerance of different populations of Korean alkali fescue,it was found that the germination rate of No.J07 material reached 58.08% with the highest germination rate under the stress-free environment.The highest relative germination potential of 213.89% was achieved under the 0.4% NaCl solution stress,indicating that the material J04 had the highest sensitivity and positive response to salt stress in the early stage of seed germination.The germination index of J01,J02,J05,J06,J07,J08,J09 and J10 all improved under low concentration salt stress conditions,and the germination rate of seeds of Korean alkali fescue could be promoted;the vigour index of J05,J06,J07,J08,J09 and J10 also improved under low concentration salt solution stress,and seed Under high salt stress,the seeds of J03,J05,J06 and J09 showed the highest germination rate and the highest salt tolerance during germination.(3)The experiment on salt tolerance in seedlings of different populations of Korean alkali fescue revealed that the changes in different indicators varied among the materials under high salt stress.The score for material J04 was 0.638,the most salt tolerant at seedling stage,and material J06 was 0.258,the least salt tolerant at seedling stage. |
