| The purpose of this study is to study the function of Mageb16 gene in the development of preimplantation embryos.This gene is a differential gene screened by transcriptome sequencing of mouse nuclear transfer embryos and normal embryos in vivo.Mageb16 gene belongs to the type I MAGE-B subfamily of the MAGE gene family,which is located on the X chromosome and encodes a protein of 363 aa.It is highly expressed in rapidly proliferating cells such as tumor cells and developing embryos,and plays a key role in the proliferation and differentiation of germ cells and tumor cells.However,the function of Mageb16 gene in preimplantation embryo development has not been reported.In this study,we used RNAi to construct a Mageb16 knockdown embryo model,and the results of in vitro culture showed that knockdown of Mageb16 resulted in lower embryo blastocyst rate,and the hatching experiment similarly showed lower embryo in vitro hatching rate.Immunofluorescence and quantitative results of lineage differentiation related marker genes showed decreased cell number,increased inner cell mass marker gene Oct4,and decreased trophectoderm Cdx2 and primitive endoderm Sox17 in the experimental group,indicating that knockdown of Mageb16 gene would lead to decreased embryonic cell number and abnormal embryonic lineage differentiation.The lack of difference between γH2AX and TUNEL assay results and the control group illustrated that the embryos did not produce obvious DNA damage and cell apoptosis,but the Ed U,p H3S10 staining results showed that knockdown of Mageb16 gene would lead to the enhanced proliferation of the embryos,but the cells were arrested in the G2/M phase,as evidenced by the elevation of p21,a cell cycle inhibitor,as quantified by the cell cycle inhibition.It is concluded that Mageb16 gene is very important for early embryonic development,especially for maintaining correct lineage norms.Moreover,Mageb16 gene can lead to abnormal early embryonic development and decreased implantation rate by inhibiting cell cycle related processes.The results showed that Mageb16 gene plays an important role in maintaining the normal development of early embryo. |
