| Osteoarthritis is a common disease worldwide,its clinical manifestations are local pain,and its main pathological features are inflammation and collagen degradation at the joints.Lactobacillus kefiranofaciens ZW3 is a probiotic that can improve the host’s intestinal microecological balance,and has anti-inflammatory and host immune-regulating effects.In view of osteoarthritis,the close relationship between gut microbiota and body inflammation has gradually been revealed,and the application of probiotics in the prevention and treatment of osteoarthritis has gradually attracted people’s attention.Therefore,this study used sodium monoiodoacetate-induced female osteoarthritis rats as the research object,and observed the effect of Lactobacillus kefiranofaciens ZW3 on osteoarthritis,which showed improved intestinal microecology and anti-inflammatory effects in previous studies.Effects of local pain,cartilage tissue pathological damage,inflammatory response and intestinal flora disturbance in rats.This study will provide a research basis for the application of probiotics in the prevention and treatment of osteoarthritis.First,in this study,the osteoarthritis model was induced by injecting sodium monoiodoacetate into the knee joint of rats,and the osteoarthritis was intervened by gavage of probiotics.The effects of Lactobacillus kefiranofaciens ZW3 on joint pain threshold and related inflammatory factors in osteoarthritis model rats were observed by Von Frey filaments and enzyme-linked immunosorbent assay.The results showed that the use of Lactobacillus kefiranofaciens ZW3 alone or in combination with chondroitin glucosamine can significantly improve the pain threshold of rats with osteoarthritis and reduce the levels of inflammatory factors IL-1β,IL-6 and TNF-α in serum.At the same time,the results also showed that the inhibitory effect of Lactobacillus kefiranofaciens ZW3 on systemic inflammation in osteoarthritis model rats can also enhance the effect of chondroitin glucosamine.Subsequently,this study further observed the pathological changes of cartilage tissue and the morphology of cartilage collagen in osteoarthritis model rats by H&E staining and toluidine blue staining.The results showed that Lactobacillus kefiranofaciens ZW3 alone and in combination with chondroitin glucosamine can effectively reduce the damage of cartilage in osteoarthritis,maintain the width and integrity of cartilage tissue,and maintain the orderly arrangement of cartilage cells.Reduce cartilage matrix decolorization.At the same time,Lactobacillus kefiranofaciens ZW3 can also decreased the expression levels of MMP1、MMP 3、 MMP13 and cyclooxygenase,and increased the expression level of type II collagen in cartilage of osteoarthritis model rats.The above results indicated that Lactobacillus kefiranofaciens ZW3 had a good protective effect on cartilage damaged in osteoarthritis.Finally,the regulatory effect of Lactobacillus kefiranofaciens ZW3 on the intestinal flora of osteoarthritis rats was analyzed by high-throughput sequencing technology.The results showed that Lactobacillus kefiranofaciens ZW3 could not only effectively improve the diversity of gut microflora destroyed in osteoarthritis,but also regulate Lactobacillus,Desulfovibrionales,Lachnospiraceae_UCG-006,and Lachnospiraceae,which are closely related to osteoarthritis.The abundance of Bacteroides and Bacteroides.In addition,the detection of short-chain fatty acids in feces showed that Lactobacillus kefiranofaciens ZW3 could also increase the content of short-chain fats that are closely related to the antiinflammatory effect produced by intestinal microbial metabolism.The results showed that Lactobacillus kefiranofaciens ZW3 had a good preventive effect on osteoarthritis by regulating the intestinal flora and its metabolites. |
