| Spermophilus dauricus is a typical fat-storing hibernator that does not eat during hibernation and relies on pre-hibernation fat as its main source of energy.From the active state to hibernation,the energy source changes from carbohydrates in the diet to stored fat,i.e.from high levels of gluconeogenesis and lipid anabolism to mainly lipolysis.In order to investigate the relationship between energy conversion of glucose and lipid metabolism during the annual cycle of the Spermophilus dauricus and the mechanisms regulating this conversion process,transcriptome sequencing and protein immunoblotting tests were performed on the livers of Spermophilus dauricus during the rapid fattening,finishing fattening and hibernation periods using Western Blotting and second-generation transcriptome sequencing(RNA-Seq)techniques,and the main results and conclusions of the analysis of differential expression of genes related to glycolipid metabolism are as follows:1.The expression of glycolytic pathway-related acetaldehyde dehydrogenase(Aldh1b1),transketolase(Tkt),aldolase bisphosphate C(Aldoc)and acyl-Co A synthetase short chain family member 2(Acss2)were significantly down-regulated in the liver at the end of the fattening period compared to the fast fattening period.fructose-bisphosphate C(Aldoc)and acyl-Co A synthetase short chain family member2(Acss2)were significantly down-regulated,and the expression of ATP citrate lyase(Acly),a gene related to the citric acid cycle,was significantly down-regulated.The expression of Acly,a gene related to the citric acid cycle,was significantly reduced,suggesting that the ability to metabolize glucose was reduced during the completion of fattening;the expression of acyl-Co A synthetase long chain family member 4(Acsl4),adipose triglyceride lipase(Pnpla3)and fatty acid binding protein 7(Fabp7)were significantly reduced,suggesting that fat synthesis was diminished at the end of the fattening period;the expression of the genes glycerol kinase(Gk),lipase The expression of glycerol kinase(Gk),phospholipase C,eta 2(Plch2)and angiopoietin like 4(Angptl4)was significantly increased,suggesting a gradual shift from anabolic to catabolic lipid metabolism.2.The expression of acetyl-Co A carboxylase alpha(Acc1),a gene related to fatty acid synthesis,was significantly decreased in the liver during hibernation compared to that of the finished fattening period,while the expression of lipase E(Lipe)and Fabp7,genes related to the metabolic pathway of lipolysis,was significantly increased,suggesting that the body is powered by lipolysis during hibernation.The results indicate that changes in material and energy metabolism in the Spermophilus dauricus precede hibernation and that hibernation is a consequence of energy stress. |
