Objective:In this study,genes related to the progression of MDS were screened out and verified by gene chip technology and bioinformatics analysis method.Meanwhile,the relationship between the expressions of RUNX1 and its key target genes and the clinical characteristics of MDS patients was further analyzed to explore their effects on the progression and prognosis of MDS.Methods:1.RUNX1 and its key target genes were screen out by cDNA microarray and bioinformatics analysis in MDS.2.Patients preliminarily diagnosed with MDS and AML were recruited from the Department of Ophthalmology of the affiliated Hospital of North Sichuan Medical College,Nanchong,China,in November 2020 to November 2021.Their bone marrow specimens from 16 MDS patients and 6 cases of AML,as well as 3 healthy controls,and all clinical data of the patients were collected.3.Total RNA was extracted using whole blood column total RNA extraction kit,and the expressions in mRNA level of RUNX1 and its key regulatory genes in bone marrow samples of MDS,AML and healthy controls were verified by real-time fluorescence quantitative PCR.4.Patients with MDS were divided into low risk group(LR-MDS)and high risk group(HR-MDS)according to international prognostic score system,and the expressions of RUNX1 and JUP in healthy control group,LR-MDS,HR-MDS and AML were compared.The correlation between RUNX1 and JUP expressions were analyzed.At the same time,the relationship between the expressions of RUNX1 and JUP and the clinical features and karyotype were analyzed.Results:1.Ten hub genes regulated by RUNX1 were screened out,namely JUP,OLFM4,CDH1,RETN,CHI3L1,PRG3,CRISP3,SLPI,MEIS1 and RHAG.2.Compared with in MDS patients,the expressions in mRNA level of JUP,RUNX1,CRISP3,MEIS1 and PRG3 in AML patients were increased,and the expression of JUP in AML was significantly higher(P<0.05).3.The results of qPCR indicated that the relative mRNA expression of JUP was gradually increased in the normal group,LR-MDS,HR-MDS and AML,and the difference was statistically significant.The relative mRNA expression of RUNX1 was also gradually increased in the corresponding group,in other words the expression levels of RUNX1 and JUP were positively correlated with risk of MDS.Compared with the lower JUP expression group,the proportion of bone marrow primitive cells in the group with high expression was significantly increased.Conclusion:The combination of the quest for disease in MDS and bioinformatics technology is beneficial to study occurrence and development of MDS.The expressions of RUNX1 and JUP are correlated with the proportion of primitive cells,platelets,and are positively correlated with the risk of MDS.JUP may be an important prognostic gene,which providing a new idea and direction for the diagnosis,treatment and prognosis evaluation of MDS.
