| Bacillus coagulans is a probiotic with spores that can produce lactic acid.It is often used in the research of producing L-lactic acid and treating intestinal diseases.This article studies the physiological and growth characteristics of Bacillus coagulans 110 stored in the laboratory,and explores the therapeutic effect and mechanism of action on inflammatory bowel disease.In this paper,through the observation of colonies and individual morphology,the size of Bacillus coagulans 110 individuals is 0.6~1.2 μm ×2.0~5.0 μm(width × length),the shape is short rod,single or paired,end sporulation,spore size 1.0 μm × 1.2 μm(width ×length),then carry out physiological and biochemical experiments according to the methods of the bacterial Berger identification manual ninth edition and GBT26428-2010,and further identify 110 strains by molecular biology methods.All results indicate that strain 110 was Bacillus coagulans.This paper studies the optimal conditions for the cultivation of 110 strains.The optimal harvesting period of Bacillus coagulans 110 is 72 h.At this time,the number of spores is about 5.98 × 106 CFU/mL.During the cultivation,the pH decreased and the acidity increased,and it stabilized at 26 h.The final pH of the bacterial solution was 4.35 and the acidity was about 11.25 g/kg.Bacillus coagulans 110 grew best in medium C at 40℃.Under this condition,the flask was cultured for 72 h,the number of viable bacteria was 1.3 × 109 CFU/mL,and the number of spores was 1.93 × 107~2.67 × 107 CFU/mL.And through the acid and bile salt tolerance experiment,it was found that Bacillus coagulans 110 has a very strong tolerance to simulated gastric acid and bile salts.In this paper,colitis mice induced by dextran sodium sulfate were used as models to study the effect of Bacillus coagulans 110 on colitis in mice.The results show that Bacillus coagulans 110 can relieve symptoms such as weight loss,blood in the stool,and damage to colon tissue in mice,and by inhibiting the relative transcription level of pro-inflammatory cytokines,promote the relative transcription level of anti-inflammatory cytokines,and adjust the immune response imbalance state.Bacillus coagulans 110 can regulate the balance of Th1/Th2 by reducing Th1 cells and increasing Th2 cells,and can also reduce the number of Th1 7 cells.Bacillus coagulans 110 inhibits the activation of the mTOR pathway by inhibiting the phosphorylation of AKT,mTOR,and p70S6K proteins,and thus it can be seen that Bacillus coagulans 110 has a therapeutic effect on colitis in mice. |
