Fusarium graminearum is the phytopathogenic fungi that caused Fusarium head blight(FHB)which is one of the most catastrophic wheat diseases worldwide.This fungus not only cause serious economic losses,its secondary metabolites also contaminate food crops and pose a great threat to the health of humans and animals.Therefore,the research about F.graminearum is highly significant to agricultural economy and food safety.Rab8 is involved in the regulation of protein transport and polarized growth by mediating the transport of vesicles from the Golgi apparatus to the plasma membrane,which plays an important role in the growth,development as well as toxin production in F.graminearum.To understand the mechanism and regulatory network of Rab8 in F.graminearum,we perfermed GFP Pull-Down combined with mass spectrometry to screen the interacting proteins of FgRab8 in F.graminearum.We obtained 696 candidate interacting proteins in total,and performed preliminary functional analysis on five candidate interacting proteins.Our results shown that FgGDTl and Fg09914 play a vital role in growth,hyphae branching,spore morphology,sexual and asexual reproduction,pathogenicity of F.graminearum.Gdt1 is mainly localized to the Golgi apparatus in yeast and in humans,regulates the transport of calcium ion,but its specific function in filamentous fungus especially in plant pathogenic filamentous fungus,remained largely unknown.In our study,FgGDTl deletion mutants shown significantly decrease in the growth rate,conidiation and pathogenicity on the wheat head,and defects in ascospore production.Furthermore,we found that Fg09914 only present in some fungi while no exist in yeast.To our knowledge,this is the first report that Fg09914 is localized to the Golgi apparatus and is required for the growth and development of F.graminearum.In addition,we found that Fg00615,Fg08543 and Fg10138 are not important to the growth conidiation and pathogenicity in F.graminearum.In summary,in this study we screened 696 candidate interacting proteins of FgRab8 in F.graminearum and selected 5 candidate proteins for futher functional characterization.We found that FgGdtl and Fg09914 were crucial for growth,development and pathogenicity in F.graminearum.
