| The first section of this study was to make a surveillance program targeting Avian Influenza Virus(AIV)in live birds in Fuzhou zoo and the periphery.A total of 361 live birds from 10 Orders,13 species in Fuzhou were examined to detect AIV.127 birds from 8 orders were found to have genes of influenza A viruses,the average positive rate is about 35%.The bird species of the highest viral prevalence rate in the zoo was Columbifirmes(53.33%),the highest viral prevalence rate in the live poultry markets was Passeriformes(46.75%).4 birds from the zoo were identified to contain genes of H9N2 subtypes,but no signals were detected with H9N2,H5 or H7 in the all samples of the live poultry from markets.The goal of the second section of this study was to make the monoclonal antibodies against H1N1 Avian Influenza.The Balb/c mice were immunized with purified A/WSN/33(H1N1)viral antigen.Then the spleen cells form immunized mice were fused with Sp2/0 myeloma cells to prepare the H1N1 specific antibodies sereted by hybridoma cell line.Two hybirdoma cell lines were generated,which could steadily secret specific monoclonal antibodies(McAbs)against H1N1 HA.These two McAbs were named as McAb-3-59 and McAb-6-32.Their HI titers were ranged from 214~215 respectively.Their neutralization test titers were ranged from 3000~4000.The results from Western blotting futher confirmed that the two McAbs could recognize the 72 KD virus structure protein indicating that they are antibodies against H1N1 HA.There were some differences between Western blotting analysis and Hemagglutination inhibition test with antigens of A/WSN/33(H1N1),PR8/(H1N1)and 09/(H1N1).The phenomenon suggested that the McAbs binding site of the these different antigens could have the different distribution.Therefore,the differences of epitome conformation of these antigens might cause the disability of the antibodies to bind after treatment with organic solvent.The animal regress experiments showed that the pretreament with the antibodies protected the animals from the virus infection,suggesting that our system may be useful for anti-influenza and the enhanced immunization and timely vaccination are important for control of this disease. |
