Purpose To observe the effect of electroacupuncture on blood sugar,body weight,SOD,CAT activity and MDA content in heart,liver,and kidney tissues of diabetic rats,and compare the three intervention methods of blank group,model group and electroacupuncture group to observe whether electroacupuncture can reduce the effect of electroacupuncture.Fasting blood sugar can improve the oxidative stress index of heart,liver and kidney,and provide experimental basis for the application of electroacupuncture in the treatment of diabetes.MethodsAfter feeding 18 8-week-old healthy male SD rats with adaptive diet for 1 week,6rats were randomly selected as blank group,and the remaining 12 rats were used as modeling group,and were fed with high-fat and high-sugar for 6 weeks.After the model was successfully induced by zotocin(STZ)30mg/kg(concentration 3.0mg/ml intraperitoneal injection)model,the diabetic rats were randomly divided into two groups: model group,electroacupuncture group,blank group and model group were not treated with Electroacupuncture,but each time it was tied in the same way as the electroacupuncture group.The electroacupuncture group was given electroacupuncture intervention.Bilateral Yishu,Housanli(Zusanli),and Sanyinjiao were taken,and the acupuncture depth was about 4 mm.The instrument selects the density wave,5Hz,3-5m A,20 min each time,and the intensity is based on the slight contraction of the muscles at the stimulation site and the tolerance of the rats,6 days a week,and a total of 4 weeks of intervention.Rats in each group were measured for body weight and tail tip blood glucose before and after modeling,and after the weekly electroacupuncture intervention.After the last intervention,the rats in each group were fasted for 12 hours.The fasting blood glucose and body weight of the rat were completely anesthetized with sodium pentobarbital,and blood was collected from the eyeball.After the blood sample was taken,the limbs of the rat were fixed on the rat plate,and a part of the heart,liver and kidney were taken and placed in liquid nitrogen for cryopreservation.For the detection of oxidative stress indicators,WST-1 method,thiobarbituric acid(TBA)method,and ammonium molybdate method were used to detect the activities of SOD and CAT and the content of MDA in three tissues.ResultBody massComparison between groups: Before the intervention,compared with the blank group,the body mass of the model group and the electro-needle group was significantly lower than that of the blank group,and the difference was statistically significant(P<0.05),and there was no significant difference between the body mass of the rats in the electro-needle group compared with the model group,and the results were not statistically significant(P>0.05).After the intervention of 4 weeks,compared with the blank group,the body mass of the rats in the model group and the electro-needle group was significantly lower than that of the rats in the blank group,and the results were very statistically significant(P<0.01),and the body mass of the rats in the electro-needle group was higher than that of the model group,and there was no obvious difference in body mass between the two groups(P>0.05).Intra-group comparison: The body mass of rats in the blank group was significantly higher than before the intervention,while the quality of the rats in the model group and the electro-needle group was reduced compared with that before and after the intervention,and the comparison between the three groups of rats before and after the intervention was significant,and the differences were statistically significant(P<0.05).Blood sugarBefore the intervention,the fasting blood glucose of the model group and the electro-needle group after long-term high-fat and high-sugar feed feeding was significantly higher than that in the blank group,and the fasting blood glucose of the three groups of rats had obvious differences,and the difference was very statistically significant(P<0.01),and the fasting blood glucose in the electro-needle group was not statistically significant compared with the model group(P>0.05);after the 4 weeks of intervention,compared with the blank group,the fasting blood glucose in the model group was significantly increased,and the difference was statistically significant(P<0.01),and the fasting blood glucose of the rats in the electro-needle group was significantly reduced compared with the model group(P<0.05).Oxidative stress index:Heart: After the end of the intervention for 4 weeks,the SOD activity,MDA content and CAT activity of the rats in the electro-acupuncture group were not statistically significant(P>0.05)compared with the blank group;the MDA content in the cardiac tissue of the rats in the model group was significantly increased,the activity of CAT was significantly reduced,and the results were statistically significant(P<0.01),and the SOD activity in the cardiac tissue was reduced,and the results were statistically significant(P<0.05).Compared with the model group,the SOD activity in the heart tissue of the electro-acupuncture group was increased,the results were statistically significant(P<0.05),the MDA content was significantly reduced,the results were significantly different,the difference was very statistically significant(P<0.01),and the CAT activity was higher than that of the model group,but there was no statistical significance compared with the two(P>0.05).Liver: After the intervention of 4 weeks,compared with the blank group,the SOD,CAT activity and MDA content in the liver tissue of the rats in the electro-acupuncture group were not statistically significant;the SOD and CAT activities in the liver tissue of the rats in the model group were significantly reduced,and the MDA content was significantly increased,and the difference was very statistically significant(P<0.01),compared with the model group,the SOD and CAT activity in the liver tissue of the electroetic group was significantly increased,and the MDA content was significantly reduced,and the results were significantly different and statistically significant(P <0.05)? Renal: After the intervention of 4 weeks,compared with the blank group,the SOD,CAT activity and MDA content in the kidney tissue of the rats in the electro-acupuncture group were not statistically significant;the SOD and CAT activities in the kidney tissue of the rats in the model group were significantly reduced,the MDA content was significantly increased,the results were significantly different,the difference was very statistically significant(P<0.01),compared with the model group,the SOD and CAT activities in the renal tissue of the electro-acupuncture group were significantly increased,the results were statistically significant(P<0.05),the MDA content was significantly reduced,there was a significant difference,and the difference was statistically significant.Conclusion1.Electroacupuncture at Zusanli,Sanyinjiao and Yishu can significantly improve blood sugar in diabetic rats2.Electroacupuncture can improve the activities of SOD,CAT and MDA in the heart,liver and kidney tissues of diabetic rats,thereby improving the level of oxidative stress in diabetic rats.
