Hepatocellular Carcinoma(HCC)is a kind of highly invasive cancer worldwide,which seriously endangers human health.Tumor stem cells with self-renewal and therapeutic resistance can promote the proliferation and metastasis of HCC cells,thus promoting the occurrence and development of liver cancer.MRNAsi is an index to measure the similarity between tumor cells and stem cells,which can be used to quantify tumor stem cells,it can be called tumor stem index.However,little attention has been paid to the research of hepatocellular carcinoma based on mrnasi.Therefore,in this paper,we used the hepatocellular carcinoma related data from the Cancer Genome Atlas(TCGA)and the mrnasi index of hepatocellular carcinoma to explore the relationship1.MRNAsi is significantly up-regulated in cancer tissues,and patients with HCC can be divided into three tumor stem cell subtypes(CSCP)by using mRNAsi related genes,and the three subtypes are named CSCP-H,CSCP-L and CSCP-M according to the index of mRNAsi.2.According to the relationship between the three subtypes and clinical manifestations,CSCP-H was significantly associated with lower survival rate and shorter relapse free survival.3.Functional enrichment analysis showed that pathways related to ribosome,spliceosome and nucleotide metabolism were more active in patients in the CSCP-H group,while pathways related to amino acid and fatty acid metabolism were less active.4.Compared with CSCP-L and CSCP-M,CSCP-H has a unique genomic pattern in somatic mutation,tumor immune cell infiltration,N6-methyladenosine and DNA methylation data.CSCP-H has higher mutation and immune cell infiltration,which may make patients with CSCP-H subtype more suitable for immunotherapy.Patients with CSCP-H had abnormal DNA methylation and abnormal expression of the m6 a gene.5.In this study,we constructed a prognosis model related to CSCP by using mRNAsi related genes.The related genes in the model can significantly divide patients into low-risk and high-risk groups,and have good prognosis characteristics,which is expected to become a potential therapeutic target for HCC patients.
