Objective:The compound method of senna leaf diarrhea,drinking water restriction,and hunger and satiety was used to prepare a mouse model of spleen deficiency constipation.The mice that were successfully modeled were treated with Zhishu Decoction,and the defecation conditions of the mice,the water content of the feces and the propulsion rate of the ink colon were observed and analyzed,and the aquaporin 3(AQP3)and cyclic adenosine monophosphate(AQP3)were detected and analyzed.c AMP),protein kinase A(PKA)protein expression levels,To investigate the intestinal motility mechanism of Citrus Aurantium Tang in the treatment of spleen deficiency constipation.Methods:First,a mouse model of spleen-deficiency constipation was prepared: Sixty SPF grade Kunming mice were divided into a blank control group of 10 and a model group of 50.Mice in the model group were given senna leaf by gavage for 7 days,during which time they had normal diet and water.On the eighth day of model building,the gavage of senna leaves was stopped,and the rat food was replaced with low-fiber feed and raw rice,during which the mice were restricted from drinking water.After 8 days,the model was successfully built.The model components were divided into: Zhizhutang high concentration group,Zhizhutang low concentration group,Zhizhutang medium concentration group,Maren soft capsule group,and model control group.Feces were collected from each group of mice and the fecal water content of each group of mice before treatment and the body weight of each group of mice were detected.After 7 days of drug intervention treatment,The fecal water content and body weight of each group of mice were measured after drug treatment,and the changes of fecal water content and body weight of each group of mice before and after treatment were compared.After the last administration,mice in all groups were fasted for 24 hours,and the next day they were gavaged with self-made ink according to 0.2ml/10 g.After taking the materials,the ink colon advancing rate of each group of mice was calculated.Part of the colon tissue of the mouse was intercepted,and the protein expression levels of c AMP,PKA and AQP3 were detected by immunohistochemistry and western blotting.Results:1.Modeling situation: On the seventh day of modeling,mice appeared weight loss,fatigue,lack of food,withered and dull coat color,increased defecation frequency,and thinner feces,which indicated that the early spleen-deficiency state of mice was successfully established.After the 15 th day of modeling,the weight of the mice in the model group did not change much,the hair was rough and lacking luster,the skin was loose,the stool was dry and hard,and the fecal particles were small.Some mice even had symptoms of rectal mucosal prolapse,which is clinically similar The symptoms of patients with spleen-deficiency constipation are similar.It can be seen that the mouse model of spleen-deficiency constipation has been successfully modeled.2.Drug treatment: After treatment with Zhizhu Tang,the defecation of mice was significantly improved,and the body weight of mice in each group also changed,among which the body weight of mice in Zhizhu Tang high concentration group and Zhizhu Tang medium concentration group had a significant increase compared with the model group(P?0.01);compared with the model group,the fecal water content of mice in the high and medium concentration Zhizhu Tang group had a significant increase(P?0.01).Compared with the model group,the rate of mouse small intestine ink advancement was significantly increased in the high and medium concentration Zhizhu Tang group(P?0.01).3.Immunohistochemical detection: After the mice were taken,the colon tissue was selected,fixed,embedded,sliced,stained,and finally observed the changes of c AMP,PKA,AQP3 in the colon.Observing the average optical density value,it was found that the contents of c AMP,PKA,and AQP3 in the high and medium concentration Zhizhu Decoction group had a significant upward trend compared with the model control group(P?0.01).4.Western blot detection: After testing,it was found that the relative expression of c AMP,PKA,and AQP3 protein in the colon of the Zhizhutang group with high and medium concentrations was significantly improved compared with the model group(P?0.01).Conclusions:1.The compound method of senna diarrhea,drinking water restriction,and hunger and fullness can successfully replicate the spleen deficiency constipation mouse model,and it is relatively stable.2.The protein expression levels of c AMP,PKA and AQP3 were decreased in the colon of mice with spleen deficiency constipation.3.Zhizhu decoction with high and medium concentration has obvious therapeutic effect on spleen-deficiency constipation,which can increase the water content of feces and accelerate the rate of colon advancement.4.Zhizhu Decoction may reduce the reabsorption of water in the colon by increasing the content of c AMP,PKA and AQP3 in the colon and improve the defecation of mice.5.Zhizhu Decoction can effectively treat spleen-deficiency constipation,and Zhizhu Decoction with high concentration and medium concentration is more effective...
