Red blood cells(RBCs)are the blood cells with the highest content in the blood,and their specific morphological changes such as too large or too small,and morphological alienation is one of the symptoms of many diseases such as blood diseases,hypertension and malignant tumors.Therefore,morphological measurement of RBCs,especially RBCs in microvessels,can provide a basis for the prevention and diagnosis of many diseases.Today,the measurement of RBCs in microvessels is mostly by means of transmission electron microscopy and angiography.These methods require to use fluorescent labels and will cause irreversible damage to cells.A phase measurement method combining simultaneous phase-shifting technology and transmission laser micro-interference technology is proposed in this paper,which not only has the advantages of good vibration resistance and good real-time performance,but also has a simple optical path,small aberrations and high spatial resolution.Therefore,this paper proposes to use this method for phase measurement and restore the appearance of RBCs in microvessels.A transmission type simultaneous phase-shifting micro-interference imaging system is designed in this paper.This system is based on Mach-Zehnder interference structure,combining micro-imaging technology and micro-polarization array-based simultaneous phase-shifting technology.The working principle of phase measurement of the system is analyzed by using Jones matrix theory.Based on the classification and comparison of phase extraction and phase unpacking algorithms,combined with the analysis of the phase characteristics of RBCs and microvessels,a RBC phase extraction algorithm based on microvascular phase subtraction is proposed.A transmission type simultaneous phase-shifting micro-interference imaging system is set up.A New Zealand rabbit RBC smear sample and a RBC sample in simulated microvessels are prepared.Through measurements by doing experiments,RBC smears with a diameter of 8 ?m and a height of 2 ?m,and the morphology of RBC samples with an inner diameter of 100 ?m in simulated microvessels are obtained.Based on the experimental results,the ability of this system to measure the morphology of RBCs in microvessels is verified,and the RBC morphology in the biomedical field is proved to have a strong application prospect and value.
