Purpose: Estrogen receptor coregulator PELP1 is closely related to the occurrence and development of a variety of cancers,but its effect on liver cancer is still unclear.siRNA was utilized to explore the roles of PELP1 playing on the proliferation,plate clone formation,migration,invasion abilityof liver cancer cells.Further study was performed to clarify its molecular mechanism on liver cancer.Methods: Hepatocellular carcinoma cell Hep G2 was selected as the research object,and three pairs of siRNA sequence and the negative control NC were used to silence the PELP1 gene.Western blot was used to test the silence efficiency and the most effective siRNA sequence was selected to perform the further experiments.Use transient transfection method to transfer PELP1-homo-3 into hepatocarcinoma cell Hep G2,and then use CCK8,plate clone formation experiment,cell scratch experiment,transwell invasion experiment,transwell migration experiment to observe the effect of PELP1 siRNA on malignant biological properties of liver cancer,and then Western blot experiments are used to explore the molecular mechanismof PELP1 in tumors.Results: The screening by Western blot experiment showed that PELP1-homo-3had the best silencing effect,so the next experiment was performed using PELP1-homo-3.The CCK8 experiment showed that the proliferation ability was significantly inhibited after PELP1 was silenced.According to the plate clone formingexperiment,the clone forming ability of liver cancer cells was significantly decreased.According to the cell scratch experiment,the transwell small chamber invasion experiment and the migration experiment,the horizontal and vertical migration and invasion ability of hepatocellular carcinoma cells are obviously inhibited.Finally,Western blot experiment was used to explore the molecular mechanism of PELP1 in liver cancer.It was revealed that PELP1 can up-regulate the expression of STAT3 signaling pathway-related genes,that is,the PELP1-homo-3transfected liver cancer cells have down-regulated the expression of PELP1 protein compared with the negative control.The protein expression levels of STAT3 and phosphorylated STAT3 were both down-regulated.Conclusion: PELP1 plays an oncogenic role on liver cancer.PELP1 silencing can down-regulate the STAT3 signal pathway to inhibit the proliferation,invasion and migration of liver cancer cells.
