Artemisia rupestris L.is characteristic Uygur medicine,a household name in Xinjiang region,widely used in clinical,with significant characteristics and unique efficacy,exactly the Uygur medicine journal records A.rupestris.has protect liver,bravery,allergy,antibacterial,scattered stasis detumescence,solution effect such as snake venom,clinically often used in the treatment of common cold,hepatitis,indigestion,urticaria,allergic disease,pharyngitis,tonsillitis,snake bites,etc.,has a good effect in the treatment of influenza and hepatitis.In order to clarify the chemical components and improve the quality control system of A.rupestris,the chemical components of A.rupestris.were qualitatively characterized:Aiming to analyze the chemical profile of A.rupestris,in-lab database was established and fragmentation pathways of primary chemical types were summarized on the basis of literature datas.Structural identification was efficiently and accurately identified the main constituents via DI-MS/MSALL and UHPLC-IT-TOF-MS,44 and 124 compounds were identified,respectively.To elucidate the effective substances action and mechanism,the chemical composition of A.rupestris in vivo was qualitatively and quantitatively characterized:1.To elucidate effective material basis and metabolic pathways of A.rupestris in vivio,in-lab database and fragmentation pathway was built based on on literature datas,124 compounds assigned in A.rupestris and extensive phase I and phase II metabolic pathways.Metabolites in plasma,urine and feces of rats were systematically characterized after oral rupestonic acid and administration of A.rupestris water ectract by LC-IT-TOF-MS.A total of 15 metabolites of rupestonic acid and 145 prototypes and metabolites were tentatively identified,including 50 in plasma,108 in urine and 47 in feces,respectively.Deglycosylation,sulfation,methylation,acetylation,hydroxylation,hydrolysis,oxidation,reduction and glucuronidation were considered as the primary metabolic pathways for the main constituents of A.rupestris.Moreover,sulfation,hydroxylation and degradation mainly occurred in plasma,sulfation and glucuronidation almost occurred in urine while most compounds found in feces were the prototypes.2.To illustrate pharmacokinetic characteristics of constituents migrating in blood,pharmacokinetic profiles of 5 drug-derived components were researched in rat after oral administration of A.rupestris.using LC-MS/MS,and concentration-time curves and pharmacokinetic parameters were obtained.This research preliminarily elucidated the chemical constituents,metabolic pathways in vivo and pharmacokinetics characteristics of components commigrating in blood of A.rupestris,which provided reference for A.rupestris efficacy material basis research,action mechanism and clinical use.
