ObjectiveThrough a cross-sectional study of patients with coronary heart disease complicated with mild to moderate depression,the related factors were analyzed to explore the distribution of TCM syndromes.Through the method of network pharmacology,the effective components,target and pathway of Qilong Tongluo Jieyu Decoction in the treatment of stable angina pectoris complicated with mild and moderate depression of CHD were explored.Therefore,it provides some reference for prevention,diagnosis and treatment of CHD complicated with mild to moderate depression,and provides theoretical basis and scientific basis for Qilong Tongluo Jieyu Decoction to treat CHD complicated with mild to moderate depression.MethodsA cross-sectional study was conducted,including 100 patients with coronary heart disease admitted to the Department of Cardiology,Dongzhimen Hospital,Beijing University of Chinese Medicine from October 2020 to February 2021.Patients'information was collected,and depression was assessed by the Hamilton Depression Scale,and angina was assessed by the Seattle Angina Scale.SPSS 26.0 was used to analyze the data.The active ingredients and target of Qilong Tongluo Jieyu Decoction were searched by TCMSP platform and related literature.The relevant targets of coronary heart disease stable angina pectoris complicated with depression were obtained through GeneCards database.Cytoscape software was used to construct the active components and potential targets of Qilong Tongluo Jieyu Decoction and the potential relationship between CHD and depression.Protein interaction analysis was conducted through the STRING database,and GO function enrichment analysis and KEGG pathway enrichment analysis were conducted on the intersection targets of the two prescription through the David database.Results1.Among the 100 CHD patients,42 patients were complicated with mild to moderate depression,and the prevalence rate of CHD complicated with mild to moderate depression was 42%.2.Multivariate analysis showed that female patients had a higher incidence of CHD combined with depression,and the difference was statistically significant(OR=4.28,P<0.05).There were significant differences in hypertension grade,time of cerebral infarction and C-reactive protein(OR=1.772,1.177,6.187,P<0.05).3.Among the 42 patients with CHD complicated with mild-to-moderate depression,there were 14 patients with Qi Stagnation and Blood Stasis,11 patients with collateral deficiency wind motion,10 patients with intercombination of phlegm and blood stasis,and 7 patients with Yin deficiency and Liver depression.4.The network pharmacological results showed that Qilong Tongluo Jieyu Decoction contained 155 active ingredients,corresponding to 7254 targets.A total of 107 common targets were identified by Venn map,and the key proteins interacting with the protein network involved STAT3,IL6,AKT1,MAPK1,CXCL8,etc.2088 biological processes were obtained by GO analysis,and 127 signaling pathways were obtained by KEGG pathway enrichment.Conclusions1.The incidence of CHD complicated with mild to moderate depression is high.CHD complicated with mild to moderate depression is correlated with women,high grade of hypertension,incidence time of cerebral infarction,and elevated CRP.2.The main syndromes of coronary heart disease complicated with mild-to-moderate depression are Qi Stagnation and Blood Stasis,and collateral deficiency wind motion.3.Qilong Tongluo Jieyu Decoction treats CHD complicated with depression through multi-channels and multi-targets,which is closely related to inflammation.
