Objective: To observe the effects of antler gum gum on hair,symptoms,body weight and testis mass in rats with oligospermia of Yang deficiency.To investigate the effect of Antler gum on sperm quality in rats with oligosthenospermia induced by Yang deficiency.To investigate the effects of Antler gum on serum T,E2,FSH and LH in rats with oligospermia of Yang deficiency.To investigate the effect of Antler gum on testicular tissue morphology and CFTR protein expression in rats with oligosthenospermia induced by Yang deficiencyMethods: Fifty male SD rats aged 8 weeks were randomly divided into normal group(n = 10)and model group(n = 40).Except for the normal group,the others were treated with adenine intragastry to make the model of oligosthenospermia with Yang deficiency.After successful modeling,40 rats in the modeling group were randomly divided into antler gum group,Yougui pill group,Zuocarnitine group and model group,with 10 rats in each group.Normal group and model group were given normal saline intragastrically,and the other groups were given corresponding drugs intragastrically for 28 days.The hair color and main symptoms of each group were observed and recorded every day.The body weight of rats in each group was detected once a week.The patients were killed 24 hours after the last drug administration.Bilateral testicles were extracted and weighed by electronic scale.the left epididymis and the tail of the epididymis were extracted and placed into 4m1 37? Deychl's solution(which needed to be preheating in advance)for incubation.The suspension droplets were fully cut into pieces with ophthalmic shears.The sperm density and activity of each group were analyzed in an automatic sperm analyzer.And the serum was separated after blood collection from the abdominal aorta by vacuum sampling.Then the contents of T,E2,FSH and LH in the serum were determined by ELISA.Two testis were taken and numbered respectively.The left side was fixed with paraformaldehyde and the right side was preserved with liquid nitrogen.HE staining technique was used to observe the repair effect of Antler gum on testicular tissue of rats with Yang deficiency and oligosthenozoospermia,and western blot technique was used to observe the effect of staghorn gum on the expression of CFTR protein in testicular tissue of rats with Yang deficiency and oligosthenozoospermia.Results: compared with the normal group,the rats in the model group were listless,curled up,had less diet,gained less weight,and even lost weight in the later stage,with coarse and yellow hair,reduced temperature sensation of limbs,loose stools,and more urine,showing a series of Yang deficiency.Compared with the model group,the hair of the antler gum group recovered luster,the mobility increased,the temperature sensation of the limbs increased,and the performance of Yang deficiency was improved.Before modeling,there was no statistical significance in the body weight of rats among all groups(P > 0.05).After administration,compared with model group,the body weight of antler gum group,Yougui pill group and Levocarnitine group was heavier(P < 0.05).Compared with the model group,the testicular mass in the antler gum group,Yougui pill group and Levocarnitine group increased significantly(P < 0.05).Compared with the normal control group,the sperm density in model group was significantly decreased(P < 0.01).Compared with model group,sperm density in Antler gum group,Yougui pill group and levocarnitine group was significantly increased;The differences were statistically significant(P <0.01).Compared with normal group,sperm motility in model group was significantly lower(P < 0.01).Compared with model group,sperm motility in Antler gum group and levocarnitine group was significantly increased,with statistically significant differences(P < 0.01).There was no significant difference in sperm density between Antler gum group,Yougui pill group and levocarnitine group(P > 0.05).Compared with blank group,serum T,E2 and LH in model group were decreased,and the difference was statistically significant(P < 0.05).FSH was increased(P <0.05).Compared with model group,serum T content was increased and FSH content was decreased in Antler gum group,Yougui pill group and levocarnitine group,with statistical significance(P < 0.05).The content of E2 increased,but there was no statistical significance(P > 0.05).There was no significant difference in LH content between model group and model group(P > 0.05).Compared with Yougui pill group and levocarnitine group,the serum contents of T and E2 were lower in Antler gum group,and the contents of FSH and LH were higher,but there was no statistical significance(P > 0.05).The testicular tissue of rats in the treatment group was repaired to a certain extent,the spermatogenic tubules were relatively full,the structure tended to be complete,all levels of spermatogenic cells were orderly,and more sperm cells could be seen in the lumen.Compared with the normal control group,the expression of CFTR protein in model group was significantly decreased(P< 0.01).Compared with model group,CFTR protein expression in antler glue group and left carnitine group was significantly increased(P < 0.05),CFTR protein expression in antler glue group was slightly decreased compared with Yougui pill,but the difference was not statistically significant(P > 0.05).Conclusion: Antler gum can effectively improve the hair,symptoms,body weight and other symptoms of Yang deficiency in rats with oligospermia of Yang deficiency,and can increase the quality of testis.Staghorn gum can effectively increase sperm density and enhance sperm motility in rats with oligoasthenospermia.Antler gum can increase the content of serum T and decrease the content of FSH in rats with oligospermia of Yang deficiency,which may be one of the mechanisms by which the effect of Antler gum on the hypothalamic-pituitary-testis axis can improve the spermatogenic function of rats.Antler gum can repair testicular tissue of adenine-induced oligospermia rats,and promote the expression of CFTR protein in testicular tissue.
