Objective: Metabolic syndrome,a high risk factor for cardiovascular and cerebrovascular events,has been proved to be linked to a variety of chronic diseases,including type 2 diabetes,cardiovascular diseases and tumors.However,the mechanism of metabolic syndrome is still unclear,the study of this disease is still full of challenges.Lian Qu,also known as Dai Zong Fang,is mainly used to treat abdominal obesity and abnormal glucose and lipid metabolism in metabolic syndrome.However,its role in metabolic syndrome is still unclear.Therefore,we planned to find the role of Lian Qu in metabolic syndrome from the point of view of multiple components corresponding to multiple targets with animal research and network pharmacology,to further explain its regulatory effect on metabolic syndrome,and to provide a theoretical basis for Lian Qu treatment of metabolic syndrome.Methods: 1)Pharmacodynamics of Lian Qu for the treatment of metabolic syndrome We induced the metabolic syndrome model in C57 mice with a high-glucose and high-fat diet.After several weeks of continued medication,we verified the weight loss effect of Lian Qu by observing the weight and fat index of the mice,verified the Lian Qu's hypoglycemic effect by observing glucose tolerance,insulin tolerance,glycosylated hemoglobin levels,and insulin levels,verified the lipid regulation effect of the Lian Qu by observing the levels of triglycerides,plasma total cholesterol,low-density lipoprotein cholesterol and high-density lipoprotein cholesterol,verified the protective effect of the Lian Qu in metabolic syndrome by observing HE staining and oil red O staining in the liver of the mice.2)Network pharmacologic analysis of Lian Qu for the treatment of metabolic syndrome We collected and analyzed the chemical components and physicochemical properties of six herbs in this drug,and screened the chemical components with OB?30% and DL?0.18 as potential active ingredients.We collected potential targets of these compounds,screened out targets related to metabolic syndrome,and selected targets with high binding ability as research objects through molecular docking technology.We analyzed the key compounds in Lian Qu by mapping the chemical-target network,analyzed the key targets for the treatment of metabolic syndrome by mapping the gene-gene interaction network,and explained the possible mechanism of Lian Qu therapy for metabolic syndrome through Gene Ontology analysis and KEGG signaling pathway analysis.Result: 1)Lian Qu reduced the body weight and the fat index of the HFD-induced C57BL/6J mice with metabolic syndrome.Lian Qu significantly improved glucose tolerance and insulin sensitivity,and decreased glycosylated hemoglobin levels in mice.Lian Qu effectively increased HDL-C and decreased the levels of TC,TG and LDL-C in serum.Lian Qu reduced the liver index of the mice,relieved the liver fatty lesions,and reduced the inflammatory infiltration in the liver.2)There are 466 compounds Lian Qu,among which 71 compounds satisfy OB?30% and DL?0.18.These compounds correspond to 292 targets,of which 74 are related to metabolic syndrome.Molecular docking results showed that 38 compounds and 45 targets had strong binding ability.Many compounds in Lian Qu can interact with multiple targets,including quercetin,luteolin,kaempferol,naringin,sesamin,some potential targets,such as AKT,TNF,IL-1?,etc.,have a high degree value in the analysis of gene-gene interaction,and these compounds and targets may play an important role in the treatment of the metabolic syndrome.The GO analysis suggested that Lian Qu therapy for metabolic syndrome may be involved in certain biological processes,including cellular response to lipid,regulation of inflammatory response,response to oxidative stress and glucose transmembrane transport.The KEGG pathway analysis results suggested that Lian Qu's treatment of metabolic syndrome may be dependent on AGE-RAGE signaling pathway,NF-?B signaling pathway and PPAR signaling pathway.Conclusion: 1)Lian Qu can effectively improve the glucose and lipid metabolism of the HFD-induced C57BL/6J mice with metabolic syndrome,and play a role in reducing blood glucose level,regulating blood lipid and losing weight.2)The multiple components in Lian Qu regulate multiple targets,participate in multiple biological processes,so as to lower blood glucose,regulate blood lipid,reduce inflammation and play a role in the treatment of metabolic syndrome.Innovative points: We verified the pharmacodynamics of Lian Qu in the treatment of metabolic syndrome in HFD-induced C57BL/6J mice with metabolic syndrome,and analyzed the mechanism of Lian Qu prevention and treatment of metabolic syndrome by network pharmacology for the first time.
