Agaricus blazei Murill was originated from Brazil,Peru and other places,which is also known as Agaricus blazei.In China,garicus blazei Murill appears mainly in Fujjian,Sichuan and Jiangsu Province.Agaricus blazei is a kind of edible and medicinal fungi which contains large amount of polysaccharides with antiviral and anticancer biological functions,therefore,the culture of Agaricus blazei will bring great profit.The purpose of this study is to explore the feasibility of a new formula culturing Agaricus blazei in Heilongjiang Province,using corncob as the main cultivation material.Then the appropriate storage temperature and method of Agaricus blazei Murill strain and wheat fungus were explored.At the same time,the polysaccharides from Agaricus blazei Murill were extracted.The following are the main methods and results:The use of cultivation material formulation for corn cob 78 %,14 % of the corn powder,white lime 3 %,the vegetation ash 2.5 %,gypsum powder 1%,diammonium phosphate 0.7 %,urea 0.6 %.With this formula,the weight of Brazil mushroom body which is produced by the fermentation material can reach 104.8±21.7g.Will Brazil mushroom and the grain of bacteria respectively at room temperature respectively after preservation for 30 d,60d and 90 d and 120 d and 0d,30 d,60d,rotating tube and inoculation.The results showed that under normal temperature,the longest preservation period of Agaricus blazei Murill is 60 d,while wheat bacteria cannot be achieved in normal temperature.Then wheat bacteria and Agaricus blazei Murill were placed in 4 ?,10 ?,15 ?,20 ?,25 incubator,removed them after 60 days ?and 120 days,by culture and observation method to explore the suitable observation Agaricus blazei Murill hatsutake bacteria and grain storage temperatures.The results showed that at 10 ?,Agaricus blazei Murill could be deposited over 120 days,while grain bacteria could deposit at least 60 d.Thus,it can be inferred that the appropriate preservative temperature was about 10 ?.In this study,by comparing the hot water extraction and enzymatic extraction,the extraction of crude polysaccharide from Agaricus blazei Murill fruiting body dry product of different sources by gradient ethanol precipitation and vacuum freeze-drying was obtained.The results showed that the content of each component was higher than that of hot water extraction by enzymatic extraction,and the content of protein and polysaccharide were the main components of each component.Proved by the experiment,the enzyme extraction combined with microwave broken extraction Agaricus blazei Murill polysaccharide nutrients is a kind of simple and effective method of manpower and material resources saving,in which no fecal culture material culture of Agaricus blazei polysaccharide extraction rate as high as7.8%.In summary,from the perspective of Agaricus blazei Murill growth and functional polysaccharide content using no fecal culture material Agaricus blazei Murill cultivation in the northern region promotion has certain feasibility.
