| Coriander(Coriandrum sativum L.)that belongs to Apiaceae Coriandrum has a long history of eating around the world and important value in both edible and medicine.In this paper,coriander cultivated in Nanjing Gaochun area has been researched about its components and biological activities of both essential oil and water extracts in the aim of providing new basis for the utilization.The essential oil of coriander seed was extracted by steam extraction and the composition and content of essential oil were determined.Result showed that the essential oil was obtained in 0.51%yield and identified by capillary GC-MS method with 18 effective volatile components for 98.2%of the total area of peaks.It contained the components of olefins,alcohols,aldehydes and esters,and the highest component is linalool(83.43%)followed by neryl acetate(3.21%),terpinene(2.85%),alpha-pinene(2.53%),(+)-2-Bornanone(1.33%),and 2,6-Octadien-l-ol,3,7-dimethyl-,(1.11%),which had significant differences in the composition and content between seed essential oil and mature stem leaves essential oil obtained before.The bacteriostatic effect of coriander seed essential oil and its effect on the vivo of Caenorhabditis elegans(C.elegans).It was uesd bacteriostatic circle method to test coriander seed essential oil bacteriostatic effect on experiental bacteria(Escherichia coli,Staphylococcus aureus,Bacillus subtilis)and fungi(Candida albicans,Penicillium and Aspergillus niger).Among them,essential oil had the most significant inhibitory effect on Escherichia coli in bacteria with diameter 24.04±1.55 mm and Aspergillus niger in fungi with diameter 29.00±4.12 mm.The result also showed that coriander seed oil could shorten the life span of nematodes,reduce the number of offspring and behaviours and increase level of reactive oxygen species,which showed inhibition of nematode life activities.Vitro antioxidant and hypoglycemic effects of coriander seed ands.The total flavonoids contents in coriander seed water extract and mature stem leaves water extract were 3%and 4.8%and then the reduction force,clearance rate of free radical and inhibition rate of alpha-glucosidase of both of them showed concentration dependence.The hydroxyl radical scavenging rates at 10 mg/mL of coriander seed and mature stem leaves water extracts were equivalent to 0.07 mg/mL and 0.13 mg/mL of Vc respectively.The DPPH free radical scavenging rates at 3 mg/mL were equivalent to 0.07 mg/mL and 0.047 mg/mL of Vc respectively.The reduction force at 1.2 mg/mL of both extracts were equivalent to 38 μg/mL and 49 μg/mL of Vc respectively.The inhibitory effect of both water extract on alpha-glucosidase were superior to acarbose,which is non-competitive inhibition.Vivo antioxidation and mechanism of coriander seed and mature stem leaves water extracts.0.1 mg/mL,1 mg/mL and 10 mg/mL of coriander seed and mature stem leaves water extracts could significantly reduce ROS accumulation level in nematodes,showing concentration dependence.What’s more,both of two water extracts could significantly improve nematodes survival rate under the PQ acute injury of oxygen stress with the minimum effective concentrations of 10 mg/mL and 0.1 mg/mL respectively.It was found that the expression of the related genes in the insulin-signaling pathway of nematodes showed a significant increase in the expression of both daf-2 and daf-16 genes.It was added with coriander seed essential oil as a bacteriostatic agent in the initial formulation of the hand sanitizer to prepare three natural essential oil hand sanitizers in three kinds of wash type and the non-washing type(water dosage form and gel type).The characteristics of homemade hand sanitizers were stable,pleasant to smell,and good experience.The main quality indicators were in line with the national standards of QB/T 2654-2013,and the bacteriostatic effect had reached the standard of industrial products. |
