China is a large country of spent mushroom substrate production and consumption.If we abandon the spent mushroom substrate at random,not only resources are wasted,but also environment will be polluted.This experiment is based on recycling the spent mushroom substrate,which consists of two points.One is alcoholic fermentation of acidolysis mushroom medium;the other is that spent mushroom substrate can be regarded as an auxiliary materials.According to mixing with corn flour in different proportions and study the quality of ethanol yield produced by mixed raw materials,alcohol production and product protein feed of alcohol production,we have achieved the following results:(1)The main components of all kinds of bacteria bran were determined.We pretreated the mushroom substrate through methods of high-temperature cooking,ultrasonic wave and microwave.By comparing the hydrolysis rate of the components cellulose,hemicellulose and lignin in the mushroom substrate.Finally the best pretreatment condition of mushroom substrate for temperature is 120 ℃,time is 2 h,and the solid-liquid ratio is 1:30.(2)We have investigated the influence of the temperature,sulfuric acid concentration,acid hydrolysis time and liquid to solid ratio which have on acidolysis efficiency.The optimum acid hydrolysis conditions of the black fungus spent mushroom substrate were optimized by orthogonal experiments.The optimized conditions were as follows: the sulfuric acid concentration is 4%,acid hydrolysis time is 1.5 h,and acidolysis temperature is 120 ℃,catalyst is phosphoric acid.(3)We measured the changes of reducing sugar concentration,sulfate ion mass concentration,acetic acid mass concentration as well as furfural mass concentration after detoxification of acid hydrolysis of spent mushroom substrate.The results show that reducing sugar concentration decreased slightly,sulfate ion mass concentration decreased obviously and most of sulfate ion were cleared up,while acetic acid mass concentration had no obvious changes before and after the detoxification.All in all,the effect of detoxification is good because spent mushroom mass concentration decreased obviously.After fermenting the ethanol yield,which was determined by adding the active yeast to the fermented solution,ultimately there is the alcoholic concentration of 0.58%.There is no meaning to use acid hydrolysis fluid of spent mushroom substrate to produce alcohol because ethyl alcohol mass concentration is low.(4)In this part,we mixed spent mushroom substrate and corn in different proportions to carry out the process of liquidation,saccharify,and alcohol production.The difference of the alcohol quality made from the mixed fermentation and pure corn fermentation was determined by GC-MS.At the same time,the quality difference of alcohol by-product distiller’s grains in crude protein,fat,crude fiber and amino acid and so on was determined.The results showed that the alcohol production made by 5% mixed spent mushroom substrate and by corn is the same.The best liquefaction p H of the mixture was 6.5,the optimum saccharification p H was 4.0,The liquefaction p H and the saccharification p H were equal to the liquefaction p H and saccharification p H of the pure corn flour.There are no significant differences in composition of the ingredients which were made by 5% mixed spent mushroom substrate and by corn.By comparing the alcohol by-product distiller’s grains made by mixed spent mushroom substrate and by corn,we found that the crude protein content is the same,the fat content is lower than that by corn,the crude fiber content is higher than that by corn,and the content of some amino acid of DDGS produced by mixed spent mushroom substrate and by corn has no obvious differences.By acid solution pretreatment and orthogonal experiment optimization conditions of acid solution,we used the acid solution fluid to produce alcohol after detoxification.Then we obtained alcohol production rate of 0.58%.This value is very low.Therefore,one hand,the ethanol production through the spent mushroom substrate need to study and develop new production technology.On the other hand,if alcohol produced by spent mushroom substrate and corn,the alcohol quality can reach the national standard.5% spent mushroom substrate can be added without any impact on the productivity of alcohol,moreover quality of DDGS is better than pure corn protein feed raw materials.
