| With China’s liquor production increased year by year,the amount of by-product distillers is increasing,but the existing treatment methods and scale can not meet the requirements.Thermop Hilic bacteria is a kind of microorganism which can grow and multiply at above 45 ℃,with the characteristics of heat thermop Hilic,degraded performance,metabolic efficiency,and it plays an important role in many areas.High-temperature heat-resistant enzyme produced by bacteria can be a good feature to speed up the degradation of lees ingredients,shorten the composting cycle significantly,the lees composting organic fertilizer,so that not only can effectively solve the problem of crop fertilizer supply,but also solve the lees resources utilization problems.In this study,separated a variety of high temperature and high temperature compost strains from the vinasse,selected seven dominant strains and studied their growth characteristics,physiological and biochemical properties and organic matter degradation,re-dubbed lees for a high temperature composting agents,and a preliminary study of high temperature composting vinasse.The main findings are as follows:(1)Different forms of more than 50 strains of thermop Hilic bacteria were screened,7 strains which were named as XX,DX,DF,M1,M4,M5 and M6 were selected for the better growth and rapid propagation at the selected temperature.Screening strains by cell morp Hology,colony morp Hology,p Hysiological and biochemical characteristics,16 S r DNA or ITS sequence analysis and p Hylogenetic analysis can initially identify the strain DX is Bacillus licheniformis、the strain XX is Bacillus aerius、the strain DF is Thermomonospora chromogena、the strain M1 is Rhizomucor pusillus、the strain M4 is Talaromyces thermop Hilus、the strain M5 is Aspergillus niger、the strain M6 is Rhizomucor tauricus.(2)The optimum growth temperature of DX and XX is 45 ℃,the cell dry weight reach the maximum between 1 to 1.5 days;DF optimum growth temperature is 55 ℃;M1 optimum growth temperature is 45 ℃,M4 optimum growth temperature is 50 ℃,the optimum growth temperature of M5 and M6 is 40 ℃,the cell dry weight reach the maximum at the third day.(3)Study on the degradation characteristics of each strain showed that DX has a strong ability to break down proteins,and its starch degradation ability also strong,the Hc value is 2.80(When the temperature is 55 ℃);the amylolytic ability of XX is strong,and XX also has some ability to decompose cellulose;DF has strong ability to decompose cellulose,which Hc value can reach 3.01(When the temperature is 55 ℃),but also has the some ability to break down starch and protein;M4 have a certain ability to break down starch and protein;M5 has strong ability to decompose cellulose,which Hc value can reach 3.21(When the temperature is 55 ℃),and has a certain amylolytic ability;M1,M6 have certain ability to decompose starch,cellulose and protein.(4)Seed germination index as the main evaluation index and the effective numberof living bacteria and E665 as secondary evaluation index,supplemented by color,odor,moisture content,p H value,electrical conductivity(EC)and other index,select group of agents J7 optimal agents,the optimal ratio of: DX: XX: M1;M4: M5: M6:DF = 1: 2: 3: 3: 0.5: 2: 6,thereby producing compost lees special compound agents,and superior to the agents in the effective number of viable cells composting and seed germination indices commercially available high-temperature agents J13 and J14.(5)Added complex agents to Lees were small-scale composting experiment,which can be concluded that vaccination composite agents can promote the pile composting fermentation,rapidly improve the composting temperature,raise the maximum reactor temperature,and prolong the time in high temperature period.The effective number of viable cells after treatment composting materials reached 760 million / g;organic matter content is greater than 40%;moisture content is reduced to less than 30%;p H rises to between 8-8.5;fecal coliform count to 3/g;roundworm eggs mortality was 99.86%;heavy metals arsenic,cadmium,lead,chromium,mercury content was 0.24,0.35,5.74,6.88,0.077mg/kg respectively;total nitrogen content increased,C and N ratio tends to 16: 1,GI> 90%,the pile full maturity,bio-organic fertilizer can meet the national standard. |
