In order to construct a suitable degradation of lignin in waste bags fungus agents.We doing the following tests were conducted.Screening strains: Acquisition nature material,then use selective media find strains can be screened for lignin degradation.Used waste bags fungus agents fermentation for a second screening.There by degrading the efficiency of the filter is relatively high strains L2 and L11 in alternative strains.Identification of strain: After screening,we select L2 and L11.Strains were identified by colony morphology,physiological and biochemical identification and molecular identification.To determine the strain L2 as Klebsiella(Klebsiella sp).The strain L11 as Bacillus(Bacillus sp).It is the basis test for subsequent genome sequencing and optimization of fermentation.Whole genome sequencing strain: the use of second-generation sequencing technology to target strains L2 and L11 were whole-genome sequencing.By sequencing analysis of the results that,L2 genome size of about 6321954 bp,predicted contain 4834 genes.L11 genome size of about 4559674 bp,predicted to contain 4131 genes.And through KEGG find some genes related to carbon metabolism pathways.Provide a reference for subsequent study the molecular mechanism of the degradation of lignin.Based on the above test results,we have been screened out two alternative strains.With further research,to explore the mechanisms of lignin degradation by bacteria and to provides a reference for the subsequent build composite agents for new high-degrading bacteria.It is also important for study the principle of lignin degradation.
