In the research of plant,male sterility is the important content of plant reproductive development.Anther as an important original place for microgametocyte,contains very complex process of male gametophyte development.Therefore,the research of the mechanism of anther and pollen development will be of great theoretical significance.Preparation of male sterile lines used for cross breeding to get heterosis is an important method of modern genetics and breeding.Thus,how to get the method of controlling male reproductive development become a key factor of creating important application value for agricultural production.In the process of anther development,tapetum cells play an important role in pollen development,bringing nutrition and necessary material to microspore.With the development and the deep research of molecular biology,people have cloned several relative transcription factors of tapetum development.However,the specific function of these genes do not have a comprehensive understanding yet.In this article,we mostly focus on MS188 and TDF1,two MYB family transcription factors on the regulatory pathway of transcription factors related to tapetum,which are fundamental purpose of this paper to analyze their function of structure and the similarity.Both of them are highly conservative in protein sequence of n-terminal,after analyzing plant phenotypic of swapping N end with C end,we found that MS188 can exercise partial function of TDF1.However,TDF1 need to add extra 44 AA sequence,which is highly conserved between species,to exercise partial function of MS188.Through the GFP subcellular localization,we approved that this 44AA could interference nuclear localization function of MS188,and verify the effect of the 44AA to activate the downstream in activation system of tobacco.The function domain of TDF1-R2R3 and MS188-R2R3 exist redundancy was determined by the EMSA experiment in vitro.These results suggest that MS188 and TDF1 as members of MYB transcription factor family exist some differences in function and molecular mechanism and some conservative features on the part of the functional domain.Summary,the results indicated that MS188 and TDF1 act as key transcription factors of anther transcriptional regulatory pathway,affects several pathways to regulate tapetum development and pollen maturation in different way.Investigation of these mechanisms deepenes our understanding of the roles of tapetal cells in pollen development.
