Cell microenvironment plays a very critical role in maintaining the normal physiological process of cells.Thanks to the development of biofluorescence imaging technology,the physiological process of the cell microenvironment that is invisible to the naked eye can be visualized through fluorescent staining.Greatly promote the research of related biological physiology and diseases.This article focuses on fluorescent probes for cell microenvironment detection.Two new types of fluorescent probes were designed for the detection of intracellular nitrosyl hydrogen(HNO)and fluorescence imaging of organelle localization.In addition,a carbon dot fluorescent probe for endoplasmic reticulum targeting was designed and synthesized using fluorescent carbon dot as material to investigate its targeting effect on endoplasmic reticulum.(1)A novel fluorescent dye matrix of bis-isophorone was designed and synthesized,using triphenylphosphine as the recognition group designing and preparing a fluorescent probe CHT-P for detecting HNO.Qualitatively and quantitatively detecting HNO and imaging of living cells were realized through the ratio signal.When the probe is combined with HNO,the fluorescence signal in the para-position of the triphenylphosphine group will be quenched,while the fluorescence signal in the ortho-position will be retained,so as to achieve the effect of HNO ratio detection.At the same time,it also shows a good ratio effect in He La cell imaging.The probe exhibits high selectivity,high sensitivity and good photostability.(2)Design and synthesize a new type of fluorescent carbon dot-based probe(CDs-PEI-HN)for highly selective fluorescence imaging of the endoplasmic reticulum.The endoplasmic reticulum localization group pentafluorobenzoic acid is used to modify and improve the fluorescent carbon dots to obtain the target probe CDs-PEI-HN.Various characterizations and spectral tests have verified the successful preparation and spectral properties of the probe.Spectral testing confirmed that the probe has the advantages of good water solubility and high light stability.Cell experiments confirmed that the fluorescent carbon dots have a better endoplasmic reticulum positioning effect.It can be further used for fluorescence imaging of the endoplasmic reticulum.Due to its high photostability,it is expected to develop into a fluorescent probe for long-term dynamic tracking of the endoplasmic reticulum of living cells.
