In population genetics,linkage analysis of genes is an important method to discover the genetic laws between population.For family members,the probability of a certain complex disease is greatly affected by genetics,so how to efficiently select as little sample sizes as possible to discover the genetic linkage in the families has become an important research direction in the field of biomedicine.The existing papers that mainly studied extreme design of close relative pairs,and demonstrated the extreme design of close relative pairs had higher power in genetic linkage analysis,but there is not much discussion on the extreme design of distant relative pairs,so focus on this problem,it has the room for further research.Based on the extreme sibpairs design,we consider extension of distant relative pairs.First,the powers and sample sizes of extreme concordant design and extreme discordant design are theoretically calculation,then we compare results of close relative pairs and distant relative pairs.The results show that for extreme discordant design,powers of two kinds of relative pairs are similar,sample sizes required to achieve 1-? power of close relative pairs are small.For extreme concordant design,powers of distant relative pairs are higher than close relative pairs,sample sizes required to achieve 1-? power of two kinds of relative pairs are similar.Finally,this paper verified through simulations research,for the extreme concordant design,when the recombination rate is small,the powers of distant relative pairs are higher than close relative pairs.But with the increase of sample size,differences of powers between two kinds of relative pairs are not obvious.For sample size of two kinds of relative pairs,only under additive model,distant relative pairs are less than close relative pairs.For dominant and recessive models,close relative pairs are less than distant relative pairs.In summary,this paper provided more sample selection strategies for linkage analysis of complex disease in families,which can improve the efficiency of linkage analysis and reduce the time costs.
