Background:Most antitumor drugs exist much deficiency such as high toxicity,poor stability and low recognition of tumor cells.Therefore,as a new type of antitumor drug sustained-release materials,injectable supramolecular hydrogels have become one of the research hotspots.Compared with polymer or macromolecular supramolecular gel systems,low molecular weight hydrogels have attracted much attention because of their high drug loading efficiency,good biocompatibility and biodegradability.However,low molecular weight hydrogels are difficult to inject because of their special physical and chemical properties and there are few reports on injectable low molecular weight gels until now.A new type of low molecular weight hydrogel was prepared by natural product isosteviol in our previous work.Objective:Based on this,novel low molecular weight injectable hydrogelator was designed and prepared by structure-optimization.After the injection of gelator precursor,the controlled-release gel system was formed in situ formation of gelator by Ion Exchange between gelator precursor and Na+in the body.Following studies on gelation properties,drug controlled-release properties and biosafety would provide useful injectable low molecular weight gels for cancer therapy and theoretical support for the design of injectable low molecular weight gels.Methods:A new type of low molecular weight gelators were synthesized by hydrolysis,esterification,sulfonation and salt formation of stevioside.Their controlled and release properties,safety,gel mechanism and properties of nano-silver preparation were studied.Result:A series of hydrogels and organogels have been synthesized with good injectability,biocompatibility and low toxicity.Self-assembly mechanism study shows that solvent molecules are involved in the self-assembly process,and the silver sulfonate-based gel system can realize the preparation of silver nanoparticles.Conclusion:The supramolecular gels have good injectability and drug controlled and release properties,and has potential theoretical and application value.Meanwhile,the silver sulfonate derivatives gel system is proved to be good template for preparing silver nanoparticles。...
