| Objective:To study the effect of carbon ion irradiation and inhibition of focal adhesion kinase expression on the biological characteristics of oral squamous carcinoma cells,and to preliminarily explore the role of focal adhesion kinase on the anti-tumor efficiency of carbon ion irradiation,providing a basis for further in vivo experiments and clinical trials.Methods: A preliminary understanding of the development status of FAK research in OSCC was performed by bibliometric analysis,and subsequent experimental was designed in combination with previous experimental results.FAK was knocked down by cell transfection to inhibit the expression of FAK protein in CAL27 cells,and the cells were irradiated with carbon ions.Cells were divided into the control sh RNA group(CK),the carbon ion irradiation group(CIR),the FAK sh RNA group(FAK-/-),the FAK sh RNA combined with irradiation group(FAK-/-+CIR).Cell proliferation was detected using colony formation assay,apoptosis was detected by a Flowsight imaging flow cytometry,protein expression was detected by Western blot,cell migration and invasion were detected by wound assay and Transwell assay,cytoskeleton was observed by immunofluorescence,Single-cell topographical and mechanical characteristics of living CAL27 cells were measured by an AFM nano-indentation method.Results: Compared with the control group,the proliferation ability of cells decreased,the number of apoptosis cells increased,the motility was inhibited,the cytoskeletal richness decreased,the cell membrane surface was rough,and viscoelasticity was weakened after carbon ion irradiation and FAK knockdown.The biological characteristics of cells treated with FAK knockdown and carbon ion irradiation were more significantly changed,the proliferation ability,migration ability and viscoelasticity were significantly decreased,and the number of apoptosis was significantly increased,membrane structure was chaged.Conclusion: The inhibitory effects of carbon ion irradiation on tumor growth and metastasis were significantly enhanced after inhibition of FAK expression,including cell proliferation ability,invasion ability,motility and viscoelasticity,partially explaining the mechanism of the anti-tumor effect of carbon ion.In addition,it provides a theoretical basis for the more effective clinical use of carbon ions in treating OSCC patients. |
