| Objective: In this project,the expression of differential protein ATAD2 before and after TSA treatment of SKBR-3 cells was screened by western-blot to verify the prophase proteomics method,and the target protein was determined.Then the expression of ATAD2 in invasive breast cancer and normal tissues was detected by immunohistochemistry,and the relationship between ATAD2 and the clinicopathological features of breast cancer was analyzed,so as to analyze the role of ATAD2 in breast cancer.Methods:(1)Cell experiment: Cells were divided into experimental group(treated by TSA)and control group,and the expression of ATAD2 protein in the two groups was detected by Western-blot and its significance was analyzed.(2)Immunohistochemistry: 60 appropriate patients with breast cancer were included,and the expression of ATAD2 protein in cancer tissues and corresponding normal breast tissues of the patients was detected by immunohistochemistry and its significance was analyzed;The clinicopathological indicators of the enrolled patients were analyzed,and the correlation between ATAD2 protein expression and pathological indicators was analyzed.Results :(1)The expression of ATAD2 in the experimental group and the control group was significantly different by western-blot assay in vitro(P < 0.05).(2)The expression of ATAD2 and Ki-67 in cancer tissues and normal breast tissues was significantly different(P < 0.05).(3)The expression of ATAD2 in breast cancer was significantly different from tumor diameter,histological grade,ER expression,breast cancer type and axillary lymph node metastasis(P < 0.05).Ki-67 expression in breast cancer patients was significantly different from tumor diameter,histological grade,PR expression and axillary lymph node metastasis(P < 0.05).There was no significant difference between ATAD2 expression and age and PR expression(P > 0.05).Ki-67 expression had no significant difference with age,ER expression and breast cancer classification(P > 0.05).(4)The expression of ATAD2 and Ki-67 was significantly different(P < 0.05);The correlation coefficient r of the two is 0.362,which can be considered as positive correlation.Conclusion: ATAD2 may plays an important role in the occurrence and development of invasive breast cancer,and is expected to become a new target for the prevention and treatment of invasive breast cancer. |
