| Forsythia suspensa is the dried fruit of Forsythia suspensa.It is widely distributed in Shanxi,Hebei and other places in China.It is a traditional Chinese medicine in China.It is known as "the holy medicine of sore family".Its main components are forsythin and forsythin a,which have many pharmacological effects such as antibacterial,anti-inflammatory,anti-virus and so on.The main contents of this subject are as follows:In this study,the fruit of Forsythia suspensa was roughly extracted with absolute ethanol,and the target compounds forsythin(5.2 g),forsythin a(244 mg),forsythin B(102 mg),forsythin D(93 mg)and forsythin e(197 mg)were isolated and purified to ensure sufficient and high purity research objects for the next research.The purity of forsythin was analyzed by NMR internal standard method.Dimethyl terephthalate was selected as the internal standard material,deuterated chloroform and deuterated DMSO as the solvent.The purity of purified forsythin is over 98.2%.Compared with HPLC,this method has high traceability and can be used as a method to determine the absolute content of reference materials.At the same time,the cellulase hydrolysis of high-purity forsythin was carried out to obtain its aglycone forsythin,which provides a material basis for the subsequent comparison of the interaction between forsythin and forsythin and human serum albumin.Through the study of the interaction between forsythin and forsythin and human serum albumin,it is found that the combination of forsythin and forsythin with human serum albumin belongs to static quenching,that is,the formation of complex leads to fluorescence quenching.Through the analysis of thermodynamic constants of forsythin and forsythin,it is concluded that forsythin has stronger binding ability,while forsythin has weaker binding ability.The main binding site of forsythin is site I,while the binding site of forsythin is site II.At the same time,for the influence of forsythin and forsythin on the secondary structure of human serum albumin,the experiment can infer that forsythin can affect the secondary structure of human serum albumin,which is specifically reflected in the decrease of α-helix value of human serum albumin skeleton after the addition of forsythin,the partial change of skeleton,the increase of hydrophobicity in the cavity of human serum albumin and the increase of the distance between the two residues emitting fluorescence.Forsythin has little effect on the structure of protein.In addition,forsythin contains sugar ring in its structure,so it has higher binding constant with human serum albumin and stronger affinity for protein.According to the comparison of binding constant data in relevant literature,it is judged that it can bind with human serum albumin in vivo,so as to achieve the purpose of transport.However,it is difficult to form hydrogen bond between forsythin structure and human serum albumin,so the binding capacity is lower,and it may not be transported with human serum albumin in the blood.Therefore,its bioavailability is low and its efficacy cannot be achieved.Forsythia suspensa and honeysuckle are used together.They have the effects of clearing heat,detoxifying and evacuating wind heat.They can be used for clearing heat and purging fire,clearing heat and purging fire,detoxifying and wind heat.The combination of the two can treat wind heat exterior syndrome,heat without sweat,slight aversion to wind cold,headache and thirst,sore throat,cough,red tip of tongue,insomnia and dreaminess.Taking honeysuckle and forsythia together can have a good curative effect on patients with symptoms such as exogenous wind heat,febrile disease,excessive anger and fire,heat entering the blood,heat toxin and so on.The combination of Forsythia suspensa and red peony has the effects of dispersing wind,clearing heat,cooling blood and relieving pain.The main treatment is just spasm.By adding chlorogenic acid,paeoniflorin and glycyrrhiza acid into human serum albumin solution,we found that these classic compatible drugs can enhance the binding ability of human serum albumin and forsythin,so as to improve the poor solubility of forsythin in blood,improve the transport capacity of forsythin,and improve the drug properties of forsythin. |
