Background:Gastric cancer remains fifth among the worldwide malignancies.During the past decade,with the comprehensive application of chemotherapy,radiotherapy,targeted therapy and other treatment strategies in clinical practice,the five-year survival rate of gastric cancer has been gradually improved.However,gastric cancer is still the third biggest cause of malignancy-related death in China.In recent years,the emergence of targeted therapy and immunotherapy has provided new insight into the treatment of advanced gastric cancer patients,such as tyrosine kinase inhibitors(TKIs),PD-1/PD-L1 monoclonal antibody,CTLA-4 monoclonal antibody,PD-1 and CTLA-4monoclonal antibody combination therapy,etc.However,due to the low efficiency and low sensitivity of the target,there is no significant clinical benefit.For example,VEGF inhibitors monotherapy did not improve the overall survival of patients with gastric cancer.Therefore,exploring new important and effective molecular markers is of far-reaching significance for improving the diagnosis rate in patients with early gastric cancer,guiding the medication of patients with advanced gastric cancer,in turn,improving the prognosis.Objective:Bioinformatics and immunohistochemical techniques were used to analyze the relationship between the m RNA expression of HDAC5 and LSD1 and the clinicopathological parameters and prognosis in patients with gastric cancer,so as to provide experimental basis for the clinical application of HDAC5 and LSD1 targets.Methods:1.We analyzed the data in online databases(Kaplan Meier Plotter database,TCGA database)to explore the relationship between the expression level of HDAC5 and LSD1 in cancer tissues and the prognosis of gastric cancer patients.2.A total of 135 patients from radical gastrectomy were obtained in the Second Hospital of Jilin University from 2010 to 2013,and immunohistochemical staining was used to analyze the differences in the expression of HDAC5 and LSD1 in gastric cancer tissues.3.Kaplan-Meier method and Log-rank test were used to analyze the prognosis of gastric cancer patients.Chi-square test was used to analyze the relationship between HDAC5 and LSD1 expression and clinicopathological parameters,and Pearson test was used to analyze the correlation between HDAC5 and LSD1 expression.4.Univariate and multivariate COX analyses were performed to analyze the relationship between the expression of HDAC5 and LSD1,the co-expression of HDAC5 and LSD1,various clinicopathological parameters and the prognosis of gastric cancer.Results:1.Using online database analysis,we found that the expression of HDAC5 and LSD1 was negatively correlated with prognosis of gastric cancer;HDAC5 expression was positively related with LSD1 expression.2.Our immunohistochemical staining showed that HDAC5 and LSD1 were overexpressed in gastric cancer tissues and verified that the expression of HDAC5 was positively correlated with LSD1 expression.3.Further univariate and multivariate Cox regression analysis has shown that the high co-expression of HDAC5 and LSD1 could be an independent risk factor for gastric cancer.Conclusion:1.HDAC5 and LSD1 were highly expressed in gastric cancer.2.The high expression of HDAC5 was positively correlated with T stage and high expression of LSD1.3.High co-expression of HDAC5 and LSD1 in gastric cancer issues may be an independent risk factor in patients with gastric cancer.
