Purpose:1.To explore the effect of drug loaded microspheres hydrogel on CRWI wound healing rate.2.To explore the effect of drug loaded microspheres hydrogel on CRWI wound collagen fibers.3.To explore the effects of drug loaded microspheres hydrogel on CRWI wound healing related proteins and genes.4.Design a drug delivery device matching the drug loaded microsphere hydrogel in this experiment.Methods:1.Establish the mouse back skin CRWI animal model by postoperative radiation;2.Establish mouse CRWI animal model,set up 3 groups:(a)experimental group(b)blank microsphere hydrogel group(c)control group.The mice were killed in batches,and the wound healing was analyzed by HE staining;The ratio of type I /III collagen was analyzed by Sirius red staining,and the distribution of collagen was analyzed by Masson staining;The expressions of VEGF-A and CD31 in wound tissue were detected by immunohistochemical staining;The expressions of healing related genes CD31,TGFB1 and VEGF-A in wound skin were detected by RT-PCR;3.Use Maya 2018 software to design a drug delivery device in line with the clinical application of this material.Results:1.Sustained-release microspheres containing leptin and vascular endothelial growth factor had good effect on promoting wound healing.2.HE staining showed that the skin tissue structure of the experimental group was more orderly,the skin accessory structures such as hair follicles recovered well,the wound tissue structure returned to normal,and the scar tissue area was smaller;3.Sirius red staining showed that in the experimental group,the proportion of type I/III collagen was lower,the area of wound scar tissue was smaller,and the arrangement of collagen fiber tissue was more orderly;4.Masson staining showed that the collagen arrangement of wound skin tissue in the experimental group was more orderly,the new collagen tissue was more and the structure was more dense;5.Immunohistochemical staining showed that compared with the control group,the number of capillaries around the wound and the expression of VEGF-A in the tissue around the wound in the experimental group increased significantly;6.RT-PCR results showed that compared with the control group,the healing related genes CD31,TGFB and VEGF-A in the wound skin tissue of the experimental group were significantly increased in the healing process;7.A syringe with a classified storage function matched with the thermosensitive drug loaded microsphere hydrogel has been developed..Conclusions:1.Thermosensitive drug loaded microsphere hydrogel has a good function in promoting wound healing after combined wound healing.2.Thermosensitive drug loaded microspheres hydrogel can promote wound healing by promoting microvascular formation and collagen secretion in wound tissue.At the same time,it can reduce the ratio of type ⅰ / ⅲ collagen in wound tissue and the area of scar tissue after wound healing;3.A syringe with a classified storage function matched with the thermosensitive drug loaded microsphere hydrogel has been developed..
