ObjectiveTo observe the effect of acupuncture on the activation state of Enteric Glial Cells in rats with visceral hypersensitivity of IBS visceral hypersensitivity animal model,Clarify whether acupuncture relieves IBS visceral hypersensitivity is related to abnormal activation of EGCs,and reveal the mechanism of acupuncture treatment of IBS visceral hypersensitivity.MethodsTwo-day-old SD rats were selected as the research object,a total of 30 rats,half male and female.The lottery method was used to randomly divide the model into a model group and a control group,of which 20 models were modeled,and the control group was 10 models.After modeling,the modeled rats were stratified and randomized according to gender and visceromotor response,and were divided into model group and acupuncture group,with 10 rats in each group.After modeling,the acupuncture group was treated with acupuncture intervention for 14 days,acupuncture Tianshu,Zusanli and Taichong points every day for 20 minutes.After the intervention,the abdominal withdrawal reflex score and the visceromotor response were used to observe their visceral sensitivity,the stool water content was used to observe their intestinal function,and the open field experiment and elevated plus maze were used to observe their anxiety and depression-like behaviors.After the behavioral evaluation,the rat colon tissue was taken for pathological examination,and the ultrastructural changes of EGC cells in the colonic submucosa were observed by transmission electron microscope.At the same time,the expression of S100 B protein in the colonic submucosa was observed by immunohistochemistry,and fluorescent double Observe the co-expression of GFAP and S100 B fluorescence using standard technology.ResultThere was no significant difference in body weight between the groups(P>0.05).Visceral sensitivity evaluation showed that the AWR score of the model group was significantly higher than that of the control group(P<0.05),and the visceromotor response of the model group was significantly lower than that of the control group(P<0.05).After acupuncture,the AWR score of the acupuncture group was significantly reduced(P<0.05),the visceromotor response was significantly increased(P<0.05).Stool conditions showed that the water content of stool in the model group was significantly increased(P<0.05),while the water content of stool in the acupuncture group was significantly reduced after acupuncture(P<0.05).Anxiety-depression-like behavior measurement showed that the model group showed obvious anxiety-depression-like state in the open field experiment and elevated plus maze experiment(P<0.05),while the acupuncture group showed a significant improvement in anxiety and depression state after acupuncture intervention(P<0.05).Colonic histopathological examination showed that there were no obvious pathological changes in colon tissues in each group,which was in line with the pathological manifestations of IBS,and this modeling method did not cause damage to rat colon tissues.The EGC morphology observation showed that the EGC cell morphology and structure of the control group were normal,while the EGC cell ultrastructure of the model group was abnormally changed,and the synaptic connection between adjacent EGC cells was increased.In addition,the EGC cell morphology and structure of the acupuncture group were normal..EGC activation state detection showed that the results of S100 B immunohistochemistry,GFAP and S100 B fluorescent double labeling showed that the fluorescence intensity of the model group was significantly higher than that of the control group(P<0.05).In addition,the expression of S100 B in the colonic submucosa of the acupuncture group was lower than that of the model Group,and the fluorescence intensity of GFAP and S100 B double-labeled fluorescence in the acupuncture group was lower than that of the model group.Conclusion1.Acupuncture can effectively alleviate the visceral hypersensitivity state of IBS visceral hypersensitivity rats,regulate gastrointestinal disorders,and reduce anxiety and depression-like behaviors.2.Acupuncture may inhibit the activation state of EGC cells and adjust the morphology and function of EGC cells,thereby alleviating IBS visceral hypersensitivity.
