Raman spectroscopy has been used more and more widely due to its molecular fingerprint characteristics and the enhancement effect of SERS.With the diversification of Raman measurement detection locations and detection methods,small portable high-throughput Raman spectrometers have more and more application prospects.In this thesis,a small Raman spectrometer is assembled by investigating various portable Raman spectrometers in the domestic and foreign markets and purchasing related accessories.And the spectrum processing algorithm is designed,the software is written for automatic control to achieve high-throughput collection operations.Finally,through various sample tests and comparative analysis with large-scale Raman microscopy,the practicability of the developed instrument was verified.The main work of this paper is as follows:1)Investigated the current development status at home and abroad,expounded the generation principle and detection mechanism of Raman spectroscopy,analyzed the internal structure and related parameters of the spectrometer,designed and developed a miniature Raman spectrometer prototype.2)In view of the current status of noise and background signals in Raman spectroscopy,the source of the noise signals are analyzed,and several different denoising algorithms are designed for MATLAB simulation,and one is finally selectd to be transplanted into the actual application software.And one is finally selected to be transplanted into the actual application software.Then analyze and apply from the perspective of software,select the de-baseline algorithm,and finally several hardware methods for remove the background signals are investigated as a supplement3)The application software is written to control it,collect data,process data,analyze data and display data,etc.This software implements the complete experimental steps from the acquisition operation to the fitting analysis operation,such as collecting data,denoising,removing background,saving read data,storing database and gradient fitting,etc.It avoids the need for the operator to change the platform operation for data analysis after collecting the data,which is more user-friendly and portable.4)Carry out systematic tests on the prototypes and software written above.The software part verifies the experimental effects of various functions,and the hardware part tests liquid samples and solid samples respectively.The solid samples include lateral flow immunoassay strips,high-throughput vertical flow immunoassay chips and porous alumina plates.The samples tested at the same time contain multiple data to verify the feasibility of multiple testing.Finally,we compared and analyzed the data tested with the Renishaw Raman Microscope,showing the advantages of high-throughput acquisition and portability of the prototype of the small Raman spectrometer developed.
