| The overall situation of domestic malignant tumor prevention and control is severe.Improving the quality of life of cancer patients and increasing their survival rate have become the main goals in the field of cancer diagnosis and treatment.The realization of this goal depends on the early detection and treatment of tumors to which the acquisition of tumor specific biomarkers is critical.Cell-SELEX(Cell-based Systematic Evolution of Ligands by Exponential Enrichment)technology can effectively select tumor specific molecular tools,aptamers which can selectively recognize tumors,thus discovery new tumor cell surface specific biomarkers which bind to aptamers.But issues including long cycle,cumbersome steps and aerosol contamination of Cell-SELEX procedure greatly restricted the discovery of tumor specific biomarkers and their detection probes--aptamers,resulting in the very limited application of this technology in the field of tumor diagnosis and treatment.Based on this,an efficient selection machine of aptamers has been developed at our research group,which can assist researchers efficiently complete the automated selection of aptamers.Based on that machine,an user-friendly,fully functional software which can realize the Cell-SELEX experimental process customization and maintain users’ experimental data has been develpoed.The main work of this paper is as follows:(1)Based on the functional design of the automated selection machine,the software requirement analysis is proposed,the development protocol is selected,and the final coding is implemented.The core business of the software is to assist users to complete the experimental process design through the graphical interactive interface,and automatically upload users’ experimental data to the cloud database;meanwhile,when the user needs and the experimental conditions permit,the software and the machine can be connected through serialport to control the operation state of the machine and track as well as display the experiment progress.(2)In order to provide users with one-stop service for experiment conduct and data research,a web-based aptamer data query system is designed and developed.The system is built into the data query page of the software through the browser environment provided by the C# Web Browser component to meet the demands of users for query,browsing and downloading of aptamers as well as the information of the aptamers related papers.(3)The functional testing and experimental verification of the developed software based on the black box testing theory are performed.The test results show that the software is smooth running,strongly interactive and highly reliable.The machine can correctly analyze the experimental process designed by the software and configure the experimental environment accordingly.The software can also correctly receive and analyze the experimental progress sent by the machine and update it on the software interface in real time.The above results show that the software developed in this study can cooperate with the machine efficiently to complete the selection of tumor specific aptamers. |
