Applicated Study Of Tumor Educated Platelet MiRNA Expression Profile In The Diagnosis Of Liver Cancer

Objective: Screen the tumor educated platelet miRNA in patients with liver cancer,find out the difference tumor educated platelet miRNA,and provide a basis for tumor educated platelet miRNA as a clinical biomarker of liver cancer.Methods: Peripheral venous blood was collected from 5 patients with liver cancer and 5 healthy adults in the same period,and platelets were separated and purified,and then sequenced using Illumina sequencing technology.Principal component analysis(PCA)method was performed on the sequencing data,and then biological information analysis was performed.Find out the differentially expressed miRNA,use the star Base database to predict each differential miRNA target,perform GO and KEGG enrichment analysis and miRNA-lnc RNA-m RNA regulatory network analysis on the differential miRNA target.Results: Principal component analysis(PCA)analysis showed that there were differences in platelet miRNA between the liver cancer group and the normal group.The gene differential expression analysis showed that compared with the normal group,there were 251 miRNA expression levels that were significantly different in the liver cancer group(including 111down-regulated miRNAs).,140 up-regulated genes).The GO enrichment analysis of differently expressed miRNA target genes suggests that the biological processes involved in down-regulation of differential miRNA targets are: intercellular signal transduction by Wnt,the positive regulation of metabolic processes,autophagy,etc.;the biological processes that up-regulated differential miRNA targets are mainly involved in are: : Positive regulation of metabolic processes,positive regulation of cell catabolic processes,forebrain development,etc.Enrichment analysis of differently expressed miRNA target genes KEGG suggests that the pathways involved in down-regulation of differential miRNA target genes are: endocytosis,proteoglycans in cancer,cell aging,etc.;the pathways involved in up-regulation of differential miRNA target genes are: Fox O signaling pathway,TGF-β signaling pathway,endocytosis,etc.The ce RNA network regulation results suggest that there are 9 meaningful network regulation pathways.Conclusion: There are differences in platelet miRNA between the liver cancer group and the normal group.Differential tumor education platelet miRNA may have certain clinical significance for the diagnosis of liver cancer.
