Objective: This study was to investigate the expression and clinical significance of sPD-1 and sPD-L1 in malignant hematological diseases by detecting the levels of sPD-1 and sPD-L1 in serum. Methods: The expression levels of sPD-1 and sPD-L1 in peripheral blood of 50 patients with newly diagnosed hematologic malignancies and30 healthy controls were detected by enzyme-linked immunosorbent assay(ELISA).At the same time,we collected a variety of serological indicators of patients and healthy people, such as white blood cell count, platelet count, hemoglobin content and other indicators, and additionally collected serum ferritin content, lactate dehydrogenase content and T cell subgroup ratio(CD4+/CD8+)of patients with hematological malignancies. To further analyze the diagnostic significance of sPD-1and spd-l1 in hematological malignancies.Results:(1)Compared with the healthy control group, the level of sPD-L1 in patients with hematological malignancies was significantly higher(P < 0.05),but there was no significant difference in the expression of sPD-1 between the two groups(P > 0.05).Further study showed that there was significant difference in the expression of sPD-1among different case groups(F = 2.795,P < 0.05),but there was no significant difference in the expression of sPD-L1 among different case groups(F = 0.549,P > 0.05).(2)Pearson correlation analysis showed that the expression of sPD-1 was positively correlated with LDH content(r = 0.235,P = 0.036),and negatively correlated with T cell subsets ratio(CD4+/CD8+)(r =-0.257,P = 0.021);the expression of sPD-L1 was positively correlated with platelet count(r = 0.307,P = 0.021).There was no significant correlation between the expression of sPD-1 and sPD-L1 and white blood cell count, hemoglobin content and ferritin content(P > 0.05).(3)The receiver operating characteristic(ROC)curve analysis showed that the sensitivity, specificity and area under the curve(AUC)of sPD-1 were 86.7%,50.0% and 0.591 respectively;the sensitivity, specificity and AUC of sPD-L1 were 80.0%,52.3% and 0.663 respectively. Conclusions: sPD-1 and sPD-L1 were correlated with disease subtypes and hematological indexes in patients with hematological malignancies, play important biological roles in hematological malignancies, and are expected to become potential biomarkers for early diagnosis and prognosis evaluation.
