Objective:To explore the effect and mechanism of Yan’s Gegen decoction on metabolic associated fatty liver disease,and provide new ideas for clinical diagnosis,treatment and new drug research and development.Methods:Thirty-eight SPF male SD rats were randomly divided into normal group(n=12)and high fat group(n=26)after adaptive feeding for 1 week.The normal group was fed with normal diet and the high fat group was fed with high fat diet for 8 weeks.Two rats in each group were randomly selected for liver histopathological examination to verify the construction of the model.After the MAFLD rat model was successfully established,the normal group was used as the control group,and the high fat group was randomly divided into model group(n=12)and prescription group(n=12).The control group was fed with normal diet,the model group and prescription group were fed with high fat diet.To study the clinical effect of Gegen decoction on MAFLD,the prescription group was intervened with Yan’s Gegen decoction for 8 weeks.Then compare the levels of triglyceride,alanine aminotransferase,aspartate aminotransferase and liver histopathological changes among the three groups after intervention.To study the mechanism of Yan’s Gegen decoction on MAFLD,the liver lipids of each group were quantitatively and qualitatively analyzed by UPLCMS/MS widely targeted liposome technology,combined with multivariate statistical analysis,differential lipids were screened and enriched by KEGG pathway.Results:1.Biochemical results: TG,ALT and AST in the model group and prescription group were higher than those in the control group,while TG,ALT and AST in the prescription group were significantly lower than those in the model group.There are statistical differences between the three groups.2.Pathological results: steatosis was not observed in the liver tissue of the control group by HE staining and oil red O staining,and there was steatosis in both the model group and the prescription group,but the degree of steatosis in the prescription group was slighter than that in the model group.3.Liver lipidomics results: 158 lipids were significantly different between the model group and the control group,105 lipids were upregulated,mainly triglyceride,cholesterol ester,etc.53 lipids were downregulated,mainly phosphatidylethanolamine,phosphatidylcholine and so on.The metabolic pathways involved include cholesterol metabolism,vitamin digestion and absorption,fat digestion and absorption and so on.There were significant differences in 9 kinds of lipids between the prescription group and the model group,in which 2 kinds of phosphatidylethanolamines were up-regulated and 7 kinds of triglycerides were down-regulated.The metabolic pathways involved include the regulation of lipolysis in adipocytes,thermogenesis,cholesterol metabolism,insulin resistance,vitamin digestion and absorption,fat digestion and absorption,and so on.Conclusions:1.Yan’s Gegen decoction has therapeutic effect on MAFLD.2.Yan’s Gegen decoction may reverse the disorder of lipid metabolism caused by high-fat diet and improve hepatocyte steatosis by down-regulating triglycerides.
