Part Ⅰ The expression levels of STIL and PRR11 and clinical significance in cervical cancer tissuesBackgroundCervical cancer poses a serious threat to women’s life and health.The occurrence and development of cervical cancer is regulated by many factors,among which the process of cell mitosis plays an important role in the proliferation and division of cervical cancer cells.STIL and PRR11 are genes that regulate mitotic centrosome replication.Previous studies have confirmed that these two proteins are over expressed in a variety of solid tumors.Over expression of STIL(SCL/TAL1 interrupting locus)and PRR11(proline-rich protein11)can promote tumor growth,but the current research in cervical cancer less.This study intends to investigate the expression of STIL and PRR11 in cervical cancer.ObjectivesTo explore the expression and clinical significance of STIL gene and PRR11 in cervical cancer tissues.MethodsA total of 67 cancer tissues and 30 cases of adjacent normal tissues from patients with cervical squamous cell carcinoma received in our hospital from May 2018 to May 2020 were selected,and the immunohistochemical method was used to detect PRR11(proline-rich protein 11)in cervical cancer tissues.And STIL gene expression to analyze the expression and clinical significance of PRR11 and STIL genes in cervical cancer tissueResultsThe results of immunohistochemistry showed that the positive signals of STIL gene and PRR11 protein were located in the cytoplasm.The positive expression rates of STIL and PRR11 were higher than those of normal tissues adjacent to cancer(P<0.05).The proportion of cases with positive STIL and PRR11 protein expression had lymph node metastasis,tumor diameter equal or larger than 4 cm,poorly differentiated tissue,and cases in the advanced stage were significantly higher than cases with negative STIL protein expression(P<0.05).The proportion of cases with double-positive STIL+PRR11 protein expression had lymph node metastasis,tumor diameter greater than 4cm,poorly differentiated tissue,and cases in the advanced stage were significantly higher than cases with double-negative STIL+PRR11 protein expression(P<0.05).ConclusionsSTIL and PRR11 are both highly expressed in cervical cancer tissues.They may be involved in the development and occurrence of cervical cancer.They are expected to become gene targets for the treatment of cervical cancer and have good clinical application value.Part Ⅱ Establishment of a prediction risk models for cervical cancer lymph node metastasisBackgroundLymph node metastasis is one of the important factors affecting the prognosis of cervical cancer.The occurrence of lymph node metastasis indicates that cervical cancer has spread.The occurrence of cervical cancer lymph node metastasis is affected by many factors,including clinical factors such as tumor size,pathological type,clinical stage,and molecular expression at the molecular biology level,but a single factor is not enough to predict the risk of cervical cancer lymph node metastasis.Therefore,based on the results of the Part I of this paper,this part plans to combine STIL and PRR11 and clinical factors to establish a nomogram model to predict the occurrence of cervical cancer lymph node metastasis.ObjectivesTo explore the risk factors that affect the lymph node metastasis of cervical cancer and establish risk prediction models.MethodsA total of 67 cases of cancerous tissues and 30 cases of adjacent normal tissues from patients with cervical squamous cell carcinoma received in our hospital from May 2018 to May 2020 were selected,and the immunohistochemical method was used to detect PRR11(proline-rich protein 11)in cervical cancer tissues.And STIL gene expression to analyze the expression and clinical significance of PRR11 and STIL genes in cervical cancer tissue.And the clinical characteristic information of the research subjects was collected.R language was performed to build a nomogram prediction model to assess the risk of cervical cancer lymph node metastasis affecting cervical cancer.ResultsThe model 1 in this study had a good predictive ability for lymph node metastasis in cervical cancer,that cervical cancer patients with large tumor size,low differentiation,and positive STIL and PRR11 expression had a significantly increased risk of lymph node metastasis.STIL and PRR11 occupied greater weight in this model.ConclusionsThe combination of differentiation degree,tumor size,poor differentitation and PRR11 expression can predict the risk of lymph node metastasis in cervical cancer.
