Objective:To study the bioactive chemical constituents of ethyl acetate extracts of Zanthoxylum nitidum(Roxb.)DC.leaves.Methods:Zanthoxylum nitidum leaves were dried naturally and crushed to powder,then the sample powders were reflux extracted four times with 70%ethanol.Each reflux were dissolved in pure water and extracted with petroleum ether,ethyl acetate and n-butanol to obtain different polar extracts.The ethyl acetate extracts were separated and purified by silica gel column chromatography,polyamide column chromatography,RP-18 column chromatography,Sephadex LH-20 column chromatography,PTLC,RPLC recrystallization and other methods.The compounds structures were elucidated by spectroscopic techniques and chemical methods,such as 1H NMR,13C NMR,ESI-MS,IR,UV.Results:Twenty compounds were isolated from the ethyl acetate extracts of Zanthoxylum nitidum(Roxb.)DC.Leaves,such as flavonoids,alkaloids,lignans,and so on.Eighteen compounds were identified,includingβ-sitosterol(1,LMYL-1),hesperetin(2,LMYL-2),ethyl caffeate(3,LMYL-4),3,4-dihydroxy benzoic acid(4,LMYL-5),4-methoxyquinolin-2-one(5,LMYL-7),pinoresinol(6,LMYL-9),medioresinol(7,LMYL-10),syringaresinol(8,LMYL-11),N-benzoyl-L-Phenylalaninol(9,LMYL-12),N-trans-coumaroyl tyramine(10,LMYL-13),L-sesamin(11,LMYL-14),hexadecanoic acid(12,LMYL-15),indolyl-3-carboxy-lic acid(13,LMYL-19),diosmetin(14,LMYL-20),zhebeiresinol(15,LMYL-25),vitexin(16,LMYL-29),isoscopletin(17,LMYL-33),loliolide(18,LMYL-6).Compounds 3,4,5,6,7,8,9,10,11,12,13,15,16,17and 18 were isolated from Zanthoxylum nitidum for the first time.Conclusions:The study provids a scientific and theoretical basis for the development and application of Zanthoxylum nitidum(Roxb.)DC.leaves.
